Melatonin is an indoleamine small molecular substance that has been shown to play an important role in the growth, development, and stress response of plants. The effects of melatonin on the morphological characteristics, mineral nutrition, nitrogen metabolism, and energy status in alfalfa (Medicago sativa L.) under high-nitrate stress were studied. The alfalfa plants were treated with water (CK), 200 mmol L−1 nitrates (HN), or 200 mmol L−1 nitrates + 0.1 mmol L−1 melatonin (HN+MT), and then were sampled for measurements on days 0 and 10, respectively. The results showed that the HN treatment resulted in a decrease in the morphological characteristics (such as shoot height, leaf length, leaf width, leaf area, and biomass), phosphorus, soluble protein (SP), nitrogen-related enzymes activities and gene relative expression, adenosine triphosphate (ATP), and energy charge (EC). It also caused an increase in nitrogen, sodium, potassium, calcium, nitrate-nitrogen (NO3--N), ammonium-nitrogen (NH4+-N), adenosine diphosphate (ADP), and adenosine monophosphate (AMP). However, these parameters were conversely changed in the HN+MT treatment. Besides, these parameters were closely related to each other, and were divided into two principal components. It reveals that melatonin plays an important role in modulating the morphology, mineral nutrition, nitrogen metabolism and energy status, thereby alleviating the adverse effects of high-nitrate stress and improving the growth of alfalfa.

Cadmium (Cd) is one of the most injurious heavy metals, affecting plant growth and development. Melatonin (N-acetyl-5-methoxytryptamine) was discovered in plants in 1995, and it is since known to act as a multifunctional molecule to alleviate abiotic and biotic stresses, especially Cd stress. Endogenously triggered or exogenously applied melatonin re-establishes the redox homeostasis by the improvement of the antioxidant defense system. It can also affect the Cd transportation and sequestration by regulating the transcripts of genes related to the major metal transport system, as well as the increase in glutathione (GSH) and phytochelatins (PCs). Melatonin activates several downstream signals, such as nitric oxide (NO), hydrogen peroxide (H2O2), and salicylic acid (SA), which are required for plant Cd tolerance. Similar to the physiological functions of NO, hydrogen sulfide (H2S) is also involved in the abiotic stress-related processes in plants. Moreover, exogenous melatonin induces H2S generation in plants under salinity or heat stress. However, the involvement of H2S action in melatonin-induced Cd tolerance is still largely unknown. In this review, we summarize the progresses in various physiological and molecular mechanisms regulated by melatonin in plants under Cd stress. The complex interactions between melatonin and H2S in acquisition of Cd stress tolerance are also discussed.

Melatonin (MT) is a signaling molecule that is widely present in plants and can help them resist abiotic stresses. In this study, the effects of different concentrations of exogenous MT on the growth and development of lettuce (Lactuca sativa) under cadmium stress (50 μmol/L) were investigated. Based on their Cd accumulation differences, the "Hong Feng" and "Luo Ma" lettuce cultivars were selected as research materials. The results showed that different concentrations of MT could promote the growth of lettuce under 50 μmol/L cadmium stress. The application of exogenous melatonin can significantly increase the plant height, biomass, chlorophyll, soluble protein and soluble sugar content of lettuce, and increase the net photosynthetic rate. MT could also reduce the malondialdehyde content in lettuce under cadmium stress and increase the activities of superoxide dismutase, peroxidase, and catalase activities. After treatment with different concentrations of MT, the Cd content in the shoots of both cultivars significantly decreased; in contrast, the Cd content in the roots was significantly increased, which limited the transport of Cd from underground to the shoots. In addition, both Cd stress and exogenous MT induced the synthesis of endogenous MT, and 50 μmol/L MT treatment most effectively induced this response. In conclusion, this study demonstrated that MT could alleviate the negative effects of Cd stress on lettuce with different Cd accumulation differences in terms of physiological indicators. Therefore, it provides a certain reference for the potential of MT to be applied to crops threatened by heavy metals.
