Exogenous melatonin alleviates cadmium uptake and toxicity in apple rootstocks

Abstract: To examine the potential roles of melatonin in Cd uptake, accumulation and detoxification in Malus plants, we exposed two different apple rootstocks varying greatly in Cd uptake and 2 accumulation to either 0 or 30 μM Cd together with 0 or 100 μM melatonin. Cd stress stimulated endogenous melatonin production to a greater extent in the Cd-tolerant M. baccata than in the Cd-susceptible M. micromalus "qingzhoulinqin". Melatonin application attenuated Cd-induced reductions in growth, photosynthesis, and enzyme ac… Show more

“…Melatonin affects biological processes not only by improving antioxidant defense systems but also by facilitating the exclusion or sequestration of heavy metals in plant cells (Hasan et al, 2015(Hasan et al, , 2019Nawaz et al, 2018;Cao et al, 2019;Sun et al, 2020). Melatonin decreases cadmium accumulation in Arabidopsis by modulating the expression of several heavy metal transporter genes, including ABC transporter genes and PCR2 (Gu et al, 2017;He et al, 2020). Taken together, these findings suggest that melatonin plays important roles in plant adaptation to heavy metal stress, functioning in a variety of ways to orchestrate this process.…”

“…The authors proposed that strigolactones act upstream of melatonin to delay flowering due to the activation of FLC when the melatonin content in tissues exceeds a certain threshold. Interestingly, NO, whose production is induced by melatonin, increases DELLA protein levels (Lozano‐Juste and León, 2011) and represses the floral transition in Arabidopsis (He et al, 2004). Shi and co‐workers (2016b), who first reported a direct link between melatonin and the floral transition, suggested that NO might be involved in melatonin‐mediated stabilization of DELLA proteins, leading to a clear delay in flowering.…”

“…It was expected that scion/rootstock interactions affected the transcription of the genes involved in Cd uptake, translocation, and detoxification. Plasma membrane-bound H + -ATPases, Fe(III) reductase, and NRAMPs regulate the uptake of divalent cations such as Cd 2+ in plants ( Zhou et al., 2016 ; Ding et al., 2017 ; He et al., 2020 ). HA7 and FRO2-like encode PM-bound H + -ATPase and Fe(III) reductase, respectively.…”

“…Our previous study found that the mRNA levels of ABCC1 and NAS1 catalyzing NA synthesis were higher in M. baccata than in M. micromalus “qingzhoulinqin”. Hence, the former had relatively greater Cd tolerance ( He et al., 2020 ). The ABCC1 , CAX4 , and MT2 transcript levels were substantially higher in Cd-treated HF/Mb than they were in Cd-challenged HF/Mm which led to comparatively higher Cd tolerance in the former.…”
