Existence of Islet Regenerating Factors within the Pancreas

Abstract: ■ AbstractReduction in the functional mass of β-cells is a common denominator in most forms of diabetes. Since the replicative potential of β-cells is limited, the search for factors that trigger islet neogenesis becomes imperative. Here we tested the hypothesis that regenerating factors for the pancreas are either secreted by or present within the pancreatic milieu itself. For this purpose, we intraperitoneally injected pancreatic cell culture supernatant (PCCS), from normal pancreas, into streptozotocin(STZ)… Show more

“…This may be due to regeneration and rejuvenation of β-cells leading to increased insulin production and secretion. These results of liver and pancreas were similar to those observed in alloxan diabetic rats subjected to plant drug therapy by earlier workers [26,27].…”

“…Then the pancreas extract solution remained in mice was intraperitoneally injected into the diabetic mice and found to reduce blood glucose levels for 190 days [28]. Kanitkar et al [29] obtained the similar results after rat pancreatic cell culture supernatant was intraperitoneally injected to diabetic rats, indicating that the proliferating pancreas can secrete a large amount of cytokines to promote regeneration of pancreatic β-cells. The injured pancreatic tissue extract promote BMMSCs differentiate into islet-like cells, was shown to significantly increase the IPCs capacity of secreting insulin [30].…”

“…Group I and Group III were administered STZ (Sigma, St Louis, MO, USA) dissolved in fresh citrate tampon (pH 4.5; 0.1 M) via intraperitoneal (IP) route in a single dose of 45 mg/kg [26] . Then, a blood sample was obtained from the tail veins of the animals following an 8-h fasting period 72 h after STZ injection and rats having a blood glucose level of 200 mg/dl or higher measured by a hand glucometer (Accu-Chek-Go, Roche, Switzerland) were considered diabetic [27] and included in the study. DEPREM, ÇINAR After the third day, when diabetes was confirmed, Group II and Group III were subcutaneously injected with capsaicin 1 mg/kg (Sigma, St Louis, MO, USA) (dissolved in 10% ethanol, 1% Tween 20, and 80% distilled water) every day for 2 weeks [28] .…”
