Exhaled eicosanoid profiles in children with atopic asthma and healthy controls

Abstract: Complementary measurements of PGEM, PGD2, LTC4, and 5HETE in small-volume EBC samples are feasible by HPLC-MS2 and showed a specific profile in our study population. EACR should be evaluated further in the context of diagnosing and monitoring childhood asthma.

“…Recently introduced methods, such as 2-dimensional protein gel electrophoresis (2D PEG) [ 57 ], followed by chromatographic proteomic microanalysis, are able to provide both qualitative and quantitative EBC-analysis. Contemporary chromatographic methods include gas chromatography (GC), high-performance liquid chromatography (HPLC), mass spectrometry (MS), and combinations of the abovementioned techniques, including LC-MS, GC-MS [ 58 ], HPLC-MS [ 59 ], Chromatography-Differential Mobility Spectrometry (GC-DMS) [ 60 ], and Electrospray Ionization-Differential Mobility Spectrometry (ESI-DMS) [ 61 ].…”

“…Likewise, increased levels of cysteinyl leukotrienes, which are powerful broncho-constrictors and pro-inflammatory mediators, have been reported in both children and adults, with the highest concentrations found in subjects with severe asthma, despite ongoing ICS therapy. 34 Several markers of oxidative stress have also been studied in EBC, pointing to an increased oxidative burst in asthmatic airways. 33 Interestingly, the simultaneous assessment of a set of metabolites of arachidonic acid led to the identification of a profile of eicosanoids (including prostaglandins and leukotrienes) capable of discriminating asthmatic and healthy children with high accuracy.…”

“…67,68 Prostaglandins and leukotrienes, both of which are arachidonic acid metabolites, have been quantified in exhaled breath condensates from children with asthma. 69 However, the extent to which asthma symptomatology and pulmonary function improve with increased intake of omega-3 or decreased intake of omega-6 fatty acids is not well known among obese children with asthma and is being investigated in ongoing randomized trials of omega-3 fatty acids supplementation. 70 Furthermore, intake of micronutrients such as vitamins A, 59, 71, 72 C, 59, 73 and E 73 has been inversely associated with asthma, whereas vitamin D insufficiency has been associated with higher asthma disease burden 74 and lower lung function.…”
