Exercise Training Promotes Cardiac Hydrogen Sulfide Biosynthesis and Mitigates Pyroptosis to Prevent High-Fat Diet-Induced Diabetic Cardiomyopathy

Abstract: Obesity increases the risk of developing diabetes and subsequently, diabetic cardiomyopathy (DMCM). Reduced cardioprotective antioxidant hydrogen sulfide (H 2 S) and increased inflammatory cell death via pyroptosis contribute to adverse cardiac remodeling and DMCM. Although exercise training (EX) has cardioprotective effects, it is unclear whether EX mitigates obesity-induced DMCM by increasing H 2 S biosynthesis and mitigating pyroptosis in the heart. C57BL6 mice were fed a high-fat diet (HFD) while undergoin… Show more

“…Performing the surgery at E11.5 also gives a good ‘therapeutic window’. The protective effect of H 2 S has been widely reported in cardiovascular [ [38] , [39] , [40] , [41] ], neurodegenerative diseases [ 42 , 43 ] and in multiple cancer pathophysiology [ [44] , [45] , [46] ]. In this study, we used MZe786, which is a H 2 S releasing aspirin as a source of H 2 S. Chronic use of aspirin is frequently associated with gastropathy, partly due to, redox imbalances [ 47 ].…”

“…NaHS treatment could improve cardiac function by targeting ROS and autophagic activities in a mouse DCM model through the elevation of Kelch-like ECH associated protein 1 (Keap-1) expression [ 187 ]. Besides, it has been shown that treadmill exercise can stabilize the decreased levels of CSE and CBS in high-fat-fed mice, thereby elevating H 2 S levels and subsequently preventing the development of DCM [ 188 ]. Pre-treatment of H9c2 cells, neonatal rat cardiomyocytes, and a mouse model with GYY4137 significantly mitigates high-glucose-induced oxidative stress and apoptosis through the promotion of FOXO-1 phosphorylation and nuclear translocation, indicating that the compound can potentially prevent the progression of DCM [ 154 ].…”

“…In addition to IF, exercise also modulates cardiovascular diseases risk factors [ 150 , 151 , 152 ]. Physical inactivity is a risk factor for cardiovascular disease and exercise is a potent preventative factor that can reduce the risk of heart disease [ 153 , 154 ].…”
