Exaggerated Spontaneous Release of Platelet-Derived Growth Factor by Alveolar Macrophages from Patients with Idiopathic Pulmonary Fibrosis

Abstract: Idiopathic pulmonary fibrosis is a fibrotic lung disease characterized by an increased number of mesenchymal cells in the alveolar walls. Alveolar macrophages constitutively express low levels of c-sis, the protooncogene coding for the B chain of platelet-derived growth factor, a protein with chemotactic and mitogenic activity toward mesenchymal cells. We therefore hypothesized that alveolar macrophages in patients with idiopathic pulmonary fibrosis may release increased amounts of platelet-derived growth fact… Show more

“…Alveolar macrophages isolated from the bronchoalveolar lavage (BAL) of patients with IPF have been shown to produce PDGF [59]. Expression of PDGF messenger ribonucleic acid (mRNA) and protein localization have also been observed directly in the lung tissue [60].…”

“…The production of these growth factors is augmented at the site of fibrosis [9][10][11], which suggests potential roles for these factors in the establishment of fibrosis. Interleukin-6 (IL-6) induces, also in vitro, fibroblast proliferation and collagen production [12,13], as well as the synthesis of tissue inhibitor of metalloproteinase-1 [14].…”

“…However, the source of platelet-derived growth factor in liver fibrogenesis is unceftain. In patients with idiopathic pulmonary fibrosis, alveolar macrophages release a large amount of platelet-derived growth factor (25). In chronic liver diseases, it is possible that activated macrophages, including Kupffer cells, and/or platelets, which accumulate in hepatic inflammation (18,26), release plateletd erived growth factor.…”
