Ex Vivo Activity Quantification in Micrometastases at the Cellular Scale Using the α-Camera Technique

Abstract: Targeted a-therapy (TAT) appears to be an ideal therapeutic technique for eliminating malignant circulating, minimal residual, or micrometastatic cells. These types of malignancies are typically infraclinical, complicating the evaluation of potential treatments. This study presents a method of ex vivo activity quantification with an a-camera device, allowing measurement of the activity taken up by tumor cells in biologic structures a few tens of microns. Methods: We examined micrometastases from a murine model… Show more

“…For these targeted strategies the anti-tumor efficacy will depend on the antigen expression and its intra-tumoral distribution i. e the targeted antigen [10, 34]. We have previously shown that, due to the short range of α-particles, the levels of absorbed radiation dose to the tumors, and other organs, could vary greatly depending on the distribution of radiolabeled antibody [18, 19]. …”

“…This has been shown on epithelial ovarian cancer (EOC) biopsies where the tumor uptake (%ID/g) radiolabeled MX35 could vary a factor of 20 in-between samples and that the activity uptake of MX35 correlated both with level and intensity of the MX35-antigen expression, as analyzed by autoradiography and immunohistochemistry [17]. Bioimaging of metastases in animal models of ovarian cancer has shown heterogenic distribution on small tumors of varying sizes [18, 19]. Strategies to predict and counteract for the impact of heterogeneity are currently being investigated, including parameters like radiation crossfire and specific activity of the radiopharmaceutical [20].…”

“…Cell death is brought about through a number of mechanisms such as apoptosis, autophagy, necrosis, and mitotic catastrophe [5357]. To insure the maintenance of the integrity of the genome, the cell is endowed with a myriad of redundant DNA repair mechanisms.…”

“…These recent techniques enable quantification of the activity distribution in tissue sections. They have been increasingly applied for research in targeted alpha radiotherapy [2528], with derivation of absorbed dose estimates [27]. In the frame of internal contamination with actinides, re-analysis of archival tissues is important to answer new questions that could not be addressed with techniques from the past [29, 30].…”
