Evolutionarily conserved function of the sacred lotus (Nelumbo nucifera Gaertn.) CER2-LIKE family in very-long-chain fatty acid elongation

Abstract: Identification of NnCER2 and NnCER2-LIKE from Nelumbo nucifera, which are required for the very-long-chain fatty acid elongation, provides new evidence that CER2 proteins are evolutionarily conserved across the eudicots. CER2-LIKE family proteins have been described as core components of the fatty acid elongase complex in Arabidopsis, maize, and rice, having specific function in synthesis of the C to C fatty acyl-CoA precursors of cuticular waxes. Little is known about the functional conservation in this gene … Show more

“…Due to its low accumulation of C 22 and C 24 VLCFAs [ 34 ], the yeast mutant strain BY4741 Δfah1 was used to investigate the function of KCS20. The CsKCS20 protein was expressed in BY4741 Δfah1 yeast, and the yeast fatty acyl chains were quantified by gas chromatography–mass spectrometry (GC–MS) ( Fig.…”

“…The coding sequence of CsKCS20 was obtained from cDNA from “Newhall” navel orange and cloned into the p426 yeast expression vector. The fatty acid methyl esters (FAMEs) of the yeast strain were analyzed as described by Yang et al [ 34 ]. Individual FAMEs were determined by the characteristic ion fragments.…”

“…A. thaliana has 21 KCS genes (Joubès et al., 2008). Characterization of the members of the KCS family showed their diverse chain‐length specificities (Yang et al., 2018). We identified KCS4 to be differentially regulated during PZ progression.…”

“…However, heterologous expression of KCS6 in yeast was found to only produce fatty acids up to 28 carbons [ 45 ]. The characterization of the CER2-LIKEs gene family in arabidopsis, but also in rice, lotus, maize or poplar, has revealed that proteins encoded by these genes physically interact with the FAE components and play a key role in specifically modifying the chain-length specificity of KCS6, enabling the production of VLCFA longer than 28 carbons [ 40 , 42 , 70 , 71 , 72 , 73 ].…”
