Evolution of Interferons and Interferon Receptors

Abstract: The earliest jawed vertebrates (Gnathostomes) would likely have had interferon (IFN) genes, since they are present in extant cartilaginous fish (sharks and rays) and bony fish (lobe-finned and ray-finned fish, the latter consisting of the chondrostei, holostei, and teleostei), as well as in tetrapods. They are thought to have evolved from a class II helical cytokine ancestor, along with the interleukin (IL)-10 cytokine family. The two rounds of whole genome duplication (WGD) that occurred between invertebrates… Show more

“…Although IFN gene families differ substantially across mammals (Secombes and Zou, 2017), the extensive expansion of IFN-u genes in R. aegyptiacus to almost two dozen genes is striking. This expansion is dramatically different from what was observed in P. alecto (Zhou et al, 2016), but is consistent with the expansion in P. vampyrus (Kepler et al, 2010).…”

“…We generated transgenic zebrafish that report on interferon signaling to visualize antiviral immune responses in live animals. Interferons are signaling proteins that induce the expression of hundreds of genes collectively termed interferon-stimulated genes, the products of which provide the main antiviral immune defenses in vertebrates (Secombes and Zou, 2017). The promoter from zebrafish interferon-stimulated gene 15 (isg15) was cloned upstream of the coding sequence for GFP in a construct that also drives constitutive GFP expression in the heart, which was inserted into the genome by Tol2 transposon-mediated transgenesis ( Figure 1A).…”

“…The coevolution of oncogenic viruses with transforming epithelial cells encourages pathogens to develop unique mechanisms that enable immune evasion. The type I interferon (IFN-I) system is an ancient and powerful host first-line antiviral defense strategy, which is conserved from human to bony fish Osteichthyes (1). Thus, viruses have to encode proteins that can block the activation of IFN-I for their replication, intercellular transmission, and/or genome integration.…”

“…HPV16 drives cancer immune escape via NLRX1-mediated degradation of STING 1 of HPV + HNSCC cancers are instead linked to HPV16 (12). More importantly, the E7 proteins encoded by HPV16 and HPV18 share a low degree of homology, suggesting potential functional divergence.…”
