Evofosfamide for the treatment of human papillomavirus-negative head and neck squamous cell carcinoma

Abstract: Evofosfamide (TH-302) is a clinical-stage hypoxia-activated prodrug of a DNA-crosslinking nitrogen mustard that has potential utility for human papillomavirus (HPV) negative head and neck squamous cell carcinoma (HNSCC), in which tumor hypoxia limits treatment outcome. Low-dose VEGF inhibitors may improve the quality of the tumor vasculature (32), elimination of hypoxic tumor cells can potentially improve response to checkpoint blockade (33), and respiratory hyperoxia can re-oxygenate the environment and improve response to immunotherapy (34). “…Patient-derived GBM cell lines (N = 13; HROG02, HROG04, HROG05, HROG06, HROG10, HROG15, HROG24, HROG36, HROG38, HROG52, HROG63, HROG73, HROG75) and HNSCC cell lines (N = 6; FADU, Detroit-562, Cal-33, PE/CA/PJ-15, UT-SCC-14, UT-SCC-15) were either established and basically characterized in our lab or originally obtained from the German collection of cell cultures (DSMZ; Braunschweig, Germany).
