We investigated the effects of incorporation of surface pre-reacted glass ionomer (S-PRG) filler in tissue conditioner (TC) on Candida albicans adhesion. We prepared specimens containing 0, 5, 10, or 20 wt% of S-PRG filler, and measured the amount of C. albicans on the surface using a colony forming unit (CFU) assay and scanning electron microscopic images. In addition, we measured the consistency, penetration depth, and surface roughness (Ra). CFU values for 10 and 20 wt% were significantly lower than that for the control (p<0.05). Hyphal density on the surface was greater in the control. The 10 and 20 wt% specimens showed significantly higher consistency and Ra, lower penetration depth ratio than control (p<0.05). These results suggest that incorporation of S-PRG filler may reduce C. albicans adhesion onto TC surface; however, the optimal amount of filler is dictated by the influence of filler incorporation on mechanical and surface characters of TC.

Statement of problem Candida-associated denture stomatitis (CADS) is the most common oral mucosal lesion among denture wearers. TMS plasma coating may inhibit the growth of Candida albicans (C. albicans) on denture surfaces. Purpose The purpose of this in vitro study was to investigate whether trimethylsilane (TMS) plasma coatings can effectively reduce C. albicans adhesion on denture base acrylic resin surfaces. Material and methods Sixty denture base acrylic resin disks (Lucitone 199) with smooth and rough surface were prepared and were either left untreated (control group) or coated with TMS monomer (experimental group) by using plasma. Contact angles were measured immediately after TMS plasma coating. The morphology of C. albicans adhesion was observed with scanning electron microscopy (SEM). Energy-dispersive spectroscopy (EDS) was used to characterize the elemental composition of the specimen surface. An adhesion test was performed by incubating the resin disk specimens in C. albicans suspensions (1×107 cell/mL) at 37°C for 24 hours and further measuring the optical density (OD) of the C. albicans using a 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assay kit. One-way ANOVA and 2-way ANOVA were followed by a post hoc test analysis (α=.05). Results The group with TMS coating exhibited a more hydrophobic surface than the control group. EDS analysis revealed successful TMS plasma coating. The difference in the mean contact angles between the uncoated group and the TMS coated group was statistically significant (P<.05), 79.0 ±2.9 degrees versus 105.7 ±1.5 degrees for the smooth surface and 90.2 ±7.6 degrees versus 131.5 ±2.1 degrees for the rough surface. In SEM analysis, the C. albicans biofilm was found to grow more on the surface of the denture base resin without the TMS coating than on the surfaces of the experimental group. In the adhesion test, the amount of C. albicans adhering to the surface of denture base resin with the TMS coating was significantly less than that on the surfaces without TMS coating (P<.05). Conclusions TMS coating significantly reduced the adhesion of C. albicans to the denture base resin and might reduce denture stomatitis.

Statement of problem:Henna has been added to polymethylmethacrylate (PMMA) as a new type of antifungal agent; however, its effect on the latter's physical properties has not been investigated. Purpose:This study aimed to evaluate the effect of the addition of various henna concentrations on the surface roughness and hardness of PMMA denture base material. Materials and Methods:A total of 99 specimens of rectangular-shaped (10×20×3 mm 3 ) acrylic specimens were prepared from heat-cured acrylic resin. Specimens were divided into one control group without the addition of henna and five test groups, which were prepared by adding Yamani henna powder to acrylic powder at concentrations of 1wt%, 2.5wt%, 5wt%, 7.5wt%, and 10wt%. The polymer was added to the monomer before being mixed, packed, and processed using the conventional water bath method. After processing, specimens were finished and polished, then kept in distilled water for 48+2 h. A profilometer and Vickers hardness tester were used to measure surface roughness and hardness respectively. Statistical data analysis was conducted via SPSS version 20.0 (IBM, USA). Results:The addition of henna at varying concentrations significantly increased surface roughness values (P ≤ 0.01) while decreasing hardness (P ≤ 0.0001). The most favorable addition value was 1% henna between all henna groups. Conclusion:The addition of henna to the acrylic resin may negatively affect the surface properties of PMMA acrylic denture base.
