Evidence of undiscovered cell regulatory mechanisms: phosphoproteins and protein kinases in mitochondria

Abstract: The finding that mitochondria contain substrates for protein kinases lead to the discovery that protein kinases are located in the mitochondria of certain tissues and species. These include pyruvate dyhydrogenase kinase, branched-chain alpha-ketoacid dehydrogenase kinase, protein kinase A, protein kinase Cdelta, stress-activated kinase and A-Raf as well as unidentified kinases. Recent evidence suggests that mitochondrial protein kinases may be involved in physiological processes such as apoptosis and steroidog… Show more

“…Cox activity is also regulated by phosphorylation and dephosphorylation. As many as at least ten of the 13 subunits of Cox could be phosphorylated at serine and/or threonine residues (Thompson 2002). As seen from our proteomics results, we found that serine/threonine-protein phosphatase 1 decreased in leucine-treated INS-1E cells (Table 1).…”

“…A number of recent studies have provided evidence that phosphorylation of mitochondrial proteins is much more frequent than expected (15)(16)(17)(18)(19)(20)(21)(22)(23). (i) Incubation of isolated mitochondria with radiolabeled ATP or staining of mitochondrial proteins with phosphospecific dyes suggested that a substantial fraction of mitochondrial proteins are phosphorylated (24 -27).…”

“…The endocrine, paracrine, autocrine, and even intracrine mechanisms that modulate this process are still to be described. No matter which one is the signaling pathway activated, the importance of the events of dynamic phosphorylation of proteins with pleiotropic effects in cellular functions as hormonal synthesis is the most relevant (Corso  Thomson, 2001;Gorostizaga et al, 2007;Thomson, 2002).…”

“…StAR protein phosphorylation mediated by PKA stimulates the transport of cholesterol from the cytoplasm into mitochondrial membranes (Stocco, 2000;Thomson, 1998;, apparently through a multiprotein complex associated to this organelle (Thomson, 2002) (Fig.10). …”
