Acute and subacute administration of zinc exert neuroprotective effects in hypoxia-ischemia animal models; yet the effect of chronic administration of zinc still remains unknown. We addressed this issue by injecting zinc at a tolerable dose (0.5 mg/kg weight, i.p.) for 14 days before common carotid artery occlusion (CCAO) in a rat. After CCAO, the level of zinc was measured by atomic absorption spectrophotometry, nitrites were determined by Griess method, lipoperoxidation was measured by Gerard-Monnier assay, and mRNA expression of 84 genes coding for cytokines, chemokines, and their receptors was measured by qRT-PCR, whereas nitrotyrosine, chemokines, and their receptors were assessed by ELISA and histopathological changes in the temporoparietal cortex-hippocampus at different time points. Long-term memory was evaluated using Morris water maze. Following CCAO, a significant increase in nitrosative stress, inflammatory chemokines/receptors, and cell death was observed after 8 h, and a 2.5-fold increase in zinc levels was detected after 7 days. Although CXCL12 and FGF2 protein levels were significantly increased, the long-term memory was impaired 12 days after reperfusion in the Zn+CCAO group. Our data suggest that the chronic administration of zinc at tolerable doses causes nitrosative stress, toxic zinc accumulation, and neuroinflammation, which might account for the neuronal death and cerebral dysfunction after CCAO.

We previously showed that the increase in nitric oxide (NO) levels and NO synthase (NOS) expression correlate with the progression of reactive astrocytosis and demyelination in the brains of 6-month-old taiep rats. Increased levels of NO can result in high concentration of peroxynitrite and thus cause tissue damage, which consists of lipoperoxidation of the cytoplasmic membrane, such as the myelin, and of apoptotic and necrotic cell-death. On this basis, we studied whether the increased NO production is associated with lipoperoxidation and cell death in the cerebellum and brainstem over the age (1, 3, 6, and 8 months) of taiep rats. The results were compared with those obtained in matched Sprague-Dawley (SD) rats. We measured the levels of nitrites (NO production), malonyldialdehyde, and 4-hydroxyalkenal (lipoperoxidation) in brain tissue homogenates. The three NOS isoforms and cleaved caspase-3 were evaluated by using ELISA and immunostaining techniques. Our results showed that NO production and lipoperoxidation increased in the cerebellum and brainstem as the age of the taiep rats increased compared to SD rats. The overexpression of nNOS and iNOS were in the Purkinje cells, magnocellular neurons, and in oligodendrocytes, whereas the glial cells showed strong cleaved-caspase-3 immunoreactivity. In summary our results suggest that NO plays a role in the demyelination and cell death in the taiep rat

The production of polyclonal antibodies (pAbs) against metallothioneins (MT) has been done in mammals. In this work, we describe a model where pAbs against rat liver MT were produced in chickens. Liver MT-1 and MT-2 isoforms isolated from rats were used as immunogens. MT was purified by exclusion chromatography and MT isoforms isolated by ionic exchange chromatography. Chickens were immunized with each isoform emulsified with Freund adjuvant over 6 weeks. MT-pAbs obtained from egg yolk were purified by ammonium sulfate precipitation followed by thiophilic interaction chromatography. MT-pAbs were characterized by ELISA, SDS-PAGE electrophoresis, and Western blot assays. Results showed significant titers (1:1,000) of MT-1 and MT-2 IgY in the eggs collected 30 days after the first immunization as determined by a direct ELISA assay; results also show a cross-reaction between MT-1 and MT-2 isoforms: however, the Abs obtained did not react with other non-MT proteins in hepatic homogenates. Sensitivity assays showed that MT-pAbs detected MT-1 and MT-2 at nanogram levels. These data suggest that chickens are an alternative model for producing pAbs against mammal high-homology proteins such as MT.
