Background: Anxiety is one the most common psychiatric disorders. Recently, anti-oxidants have been shown to possess anxiolytic properties. Objectives: The present study aimed to investigate anxiolytic effect of ellagic acid (EA) in mice and its interaction with GABAA receptor. Materials and Methods: 184 male albino mice (25 -30 g) were randomly assigned into two subgroups (12 groups in each). In the first set of experiments, to evaluate acute administration of EA (single dose) on anxiety, experimental groups (10 groups, 8 mice per group) were control (received vehicle), EA-treated groups (received ellagic acid, 3, 10, 30 and 100 mg/kg, i.p.), diazepam (DZP)-treated groups (received DZP, 1, 3 and 5 mg/kg, i.p.), Flumazenil (FLZ) + diazepam-treated group (received FLZ, 3 mg/kg and + DPZ, 5 mg/kg, i.p.) and FLZ+EA-treated group (received FLZ, 3 mg/kg and ellagic acid, 30 mg/kg, i.p.). Moreover, in the first set of experiments, to evaluate the effect of chronic administration of EA on anxiety, experimental groups (2 groups, 8 mice per group) were control (received vehicle once a day for 10 days) and EA-treated group (received ellagic acid at the dose of 30 mg/kg, i.p. once a day for 10 days). In the second set of experiments, to evaluate acute (single dose) and chronic (for 10 days) administration of EA on motor activity, animal groupings were similar to the first set of experiment. Elevated plus maze (EPM) and open field tests used to study anxiolytic and motor activity effect of EA, respectively. Statistical analysis was performed by one-way ANOVA and post hoc Tukey's test. P values less than 0.05 were considered as significant. Ellagic acid at the dose of 100 mg/kg significantly decreased ambulatory movement compared with the control, while EA at the dose of 30 mg/kg did not affect ambulation. Conclusions: Acute and chronic administration of ellagic acid had anxiolytic property. Also, the most effective dose of ellagic acid was 30 mg/kg. Pretreatment with flumazenil reversed the beneficial effect of ellagic acid and diazepam on anxiety. It is likely that the anxiolytic effect of ellagic acid is largely mediated by activation of GABAA receptor.

Alzheimer's disease is a severe neurodegenerative disorder that gradually results in loss of memory and impairment of cognitive functions in the elderly.Thedeposition of amyloid plaques is the primary event that leads to an oxidative and inflammatory reactions, neurofibrillary tangle formation, and ultimately neuronal death. The most prominent losses occur with cholinergic neurons that plays an important role in the formation of memory and cognitive functions. Insulin receptor numbers also altered, thusglucosemetabolism is defected in AD. Polyphenol ellagic acid (EA) possesses antioxidant, anti-inflammatory, anti-carcinogenic, anti-diabetic and cardioprotection activities.Activities of EA that are linked with protection of neuronal abnormalities like anti-depressant, antianxiety, anti-nociception. Ellagic acid inhibits β-secretase enzymatic activities, thus prevents the main pathologic hallmark of AD. There are varieties of herbal extracts and phytochemicals formulations extensively used to provide symptomatic relief to AD patients. Together these results our hypothesis that EA may prevent the neuronal dysfunctions as well as further work is also required to examine the pharmacological properties of EA in the protection of specially neurodegenerative disorder like AD. Here, we discuss how we can use the knowledge to improve treatment strategies of EA in AD and to explore the various signaling pathways involved in the progression of neuronal death.

Objective: The present study was initiated to describe the gastroprotective role of quercetin (Qu) and ellagic acid (EA) on aspirin-induced gastric ulcer (GU) in rats.Methods: Forty adult female albino rats of Wistar strain were distributed into: Control group, GU group, Omeprazole group, Qu group, and EA group. Gross examination, biochemical analyses including serum adrenocorticotropic hormone (ACTH), serotonin (ST), ferritin, heme oxygenase-1 (HO-1), interleukin-2 (IL-2), advanced glycosylation end products (AGEs), and fibronectin (FN) levels were estimated. Moreover, histopathological and histochemical examinations of stomach tissue samples were carried out.Results: Gross examination of gastric mucosa of rats in GU group revealed hyperemia of the stomach mucosa. Furthermore, rats in GU group experienced a significant rise in serum ACTH, ferritin, HO-1, IL-2 and AGEs levels accompanied with significant drop in serum ST and FN levels versus control counterparts. Pre-treatment of GU group with Omeprazole, Qu or EA caused marked improvement in the measured biochemical parameters. Histopathological and histochemical examinations of stomach tissue samples documented the protective action of Omeprazole, Qu and EA with different degrees against GU caused by aspirin. Conclusion:As a conclusion to this study, we can state that both Qu and EA have gastroprotective effect against aspirin-induced GU in rat model. Of note, Qu showed superior impact than EA as an antiulcer agent in this study. The corresponding mechanisms are speculated to be associated with inhibiting stress-induced gastric lesion, attenuating the oxidative stress, iron chelation and blunting ferritin level, modulating inflammatory cascade, and promoting the healing process.
