Evidence for the interaction of d-amino acid oxidase with pLG72 in a glial cell line

Abstract: Accumulating genetic evidence indicates that the primate-specific gene locus G72/G30 is related to schizophrenia: it encodes for the protein pLG72, whose function is still the subject of controversy. We recently demonstrated that pLG72 negatively affects the activity of human d-amino acid oxidase (hDAAO, also related to schizophrenia susceptibility), which in neurons and (predominantly) in glia is expected to catabolize the neuromodulator d-serine. The d-serine regulation mechanism relying on hDAAO-pLG72 inter… Show more

“…The principle on which the proposed assay is based is that pLG72 binds to hDAAO, 5,7,8 alters hDAAO structure in a time-dependent manner, 7,22 and regulates hDAAO activity. This regulation has been reported as activation 5,23 by some researchers and inhibition by others.…”

“…Some findings indicated that LG72 binds to the D-serine degrading enzyme DAO (Chang et al 2013;Chumakov et al 2002;Sacchi et al 2008;Sacchi et al 2011). However, DAO and LG72 are localized in different subcellular compartments.…”

“…In contrast, several studies have shown a mitochondrial localization of LG72 (Kvajo et al 2008;Otte et al 2011;Sacchi et al 2011). In order to reconcile the disparate sub-cellular localization with a putative modulation of DAO activity by LG72, it has been suggested that the two proteins interact in the cytosol during the transport of DAO into peroxisomes (Sacchi et al 2011). Even if this were correct, we find it highly unlikely that modulation of DAO activity is the only function of LG72.…”

“…All experiments were performed using a U87 human glioblastoma cell line stably expressing human DAAO [25]. Cellular amino acids were extracted as described previously [18]: 2.5 9 10 5 U87 cells were resuspended in 1 mL of ice-cold 5% trichloroacetic acid, sonicated and centrifuged for 45 min at 16 000 g. For the HPLC assay, the soluble fraction was extracted with water-saturated ether, neutralized and derivatized with o-phthaldialdehyde/N-acetyl-L-cysteine.…”

“…The novel sensing assay shows a detection limit ≀ 0.5 lM (slightly lower than commercial assay kits) and was used to assay glycine and sarcosine in biological fluid, i.e. in a U87 glioblastoma cell line used to study the modulation of D-serine level by the catabolic enzyme D-amino acid oxidase [18,25]; D-serine and glycine are alternative ligands required, together with glutamate, to activate NMDARs [21,26]. The glycine concentration detected by the H244K GO-based assay was in good agreement with the value obtained by using the reference HPLC method (7.5 versus 6.7 lM, respectively) but in 1 s, compared with the 60 min required by chromatographic separation.…”
