Evidence for Interleukin‐10‐Mediated Inhibition of Cyclo‐ oxygenase‐2 Expression and Prostaglandin Production in Preterm Human Placenta

Abstract: These results demonstrate the importance of IL-10 in countering inflammation associated with preterm labor, and suggest that term and preterm parturition may, in part, represent different conditions.

“…Indeed, previous studies focusing on gestational tissues have reported either no significant differences [59,62,183] or a significant reduction in IL-10 expression in the presence of labor when compared to controls [184][185][186]. Of note, Hanna et al [59] reported a labor-associated up-regulation of TNF-a and IL-1 expression in placental explants and freshly isolated cytotrophoblasts in contrast to an absence of appreciable changes in IL-10 expression [59].…”

“…Importantly, the pregnancy outcome in treated IL-10 Ϫ/Ϫ mice can be rescued by giving an exogenous dose of IL-10 (11,14). We have also demonstrated poor IL-10 production in placental and decidual tissues from preterm labor deliveries and missed abortions (15,16). These data suggest that an inflammatory environment coupled with genetic stress (IL-10 deficiency) may lead to adverse pregnancy outcomes.…”

“…Labor is associated with increased production of proinflammatory mediators such as interferon-␥ and TNF-␣, and decreased release of IL-10, a potent anti-inflammatory cytokine, and these changes are thought to play a role in the onset of parturition (18). Proinflammatory mediators have also been shown to promote neutrophil activation and longevity (4).…”
