Evidence for extrachromosomal elements in Lactobacillus

Abstract: Three strains of lactobacilli, Lactobacillus casei subsp. casei 64H, L. casei subsp. rhamnosus OC91, and L. coryniformis M34, were examined for the presence of plasmids. Plasmids of molecular weights of 23 x 10(6) and 16 x 10(6) were found in the first two strains respectively. This represents the first evidence for plasmids in lactobacilli; their function is not presently known.

“…To isolate DNA from lactobacilli, 5 ml of an overnight culture of each strain grown in MRS broth were diluted with 10 ml pre-warmed MRS broth. After about 4 h of incubation at 37°C, the bacterial cells were harvested by centrifugation and the chromosomal DNA was isolated as described by Chassy et al (1976).…”

“…Plsamid DNA was isolated from E. coli strains by a modification of the method of Birnboim and Doly [15,16]. DNA was isolated from L. casei strains by an adaptation of the method of Currier and Nester [17,18]. PL-1 phage DNA was isolated from hightitre lysates as previously described [14].…”

“…The presence of plasmids within the species of Lactobacillus has been reported since they were first discovered in this genus (Chassy et al 1976). Phenotypes such as bacteriocin production and immunity (Muriana & Klaenhammer 1987;Schillinger & Liicke 1989), lactose metabolism (Chassy et al 1978) and drug resistance (Ishiwa & Iwata 1980; Vescovo et al. 1982; Morelli et al.…”
