Evidence for an evolutionary conserved role ofhomothorax/Meis1/2during vertebrate retina development

Abstract: During eye development in D. melanogaster, the TALE-homeodomain protein Homothorax (Hth) is expressed by progenitor cells ahead of the neurogenic wave front, promotes rapid proliferation of these cells and is downregulated before cells exit the cell cycle and differentiate. Here, we present evidence that hth function is partially conserved in vertebrates. Retinal progenitor cells (RPCs) in chicks and mice express two Hth-related proteins, Meis1 and Meis2 (Mrg1), in species-specific temporal sequences. Meis1 ma… Show more

“…This regulatory cascade is strongly supported by a number of observations. Meis2 directly activates Pax6 expression during lens and retina development in the zebrafish, chick, and mouse (16,19,30). In medaka fish embryos, expression levels of olMeis2 and, concomitantly, those of olPax6 and its downstream targets are up-regulated on miR-204 knock-down or inhibition of the miR-204 interaction with its specific target site in the Meis2 3′-UTR.…”

“…cyclinD1 and myca are regulated by meis1 in the chick and zebrafish retina (Bessa et al, 2008;Heine et al, 2008). meis1 is a homeodomain transcription factor that is expressed in retinal progenitor cells in Drosophila and zebrafish and keeps them in a proliferative state and suppresses differentiation (Pai et al, 1998;Bessa et al, 2008;Heine et al, 2008).…”

“…The distinct cell-fate potentials of ventral midbrain progenitors are defined by domain-identifiable expression of transcription factors. For example, the Lmx1a + Foxa2 + FP exclusively gives rise to mDA neurons, whereas the ventral-lateral domains marked by Meis2, Mab21l2, Helt, and Lhx1 produce glutamatergic or GABAergic neurons (6)(7)(8)(9). Perturbation of such a transcription "code" seen in genetic studies often led to misspecification of progenitor identity and subsequently to neural transmitter phenotypes (10)(11)(12).…”
