Evaluation of α,β-unsaturated ketone-based probes for papain-family cysteine proteases

Abstract: The field of activity based proteomics makes use of small molecule active site probes to monitor distinct subsets of enzymatic proteins. While a number of reactive functional groups have been applied to activity based probes (ABPs) that target diverse families of proteases, there remains a continual need for further evaluation of new probe scaffolds and reactive functional groups for use in ABPs. In this study we evaluate the utility of the α, β-unsaturated ketone reactive group for use in ABPs targeting the p… Show more

“…Table 1 lists an overview of protease-reactive warheads. [24,28,39,40] Serine protease warheads have been based on fluorophosphonates, [8,38] diphenyl phosphonates, [17][18][19] and isocoumarins. The sulfur atom in the active site of a cysteine protease is softer and more nucleophilic than the oxygen atom in a serine protease.…”

“…Therefore, selective targeting of cysteine proteases is possible through the use of soft electrophiles such as acyloxymethyl ketones, [16] epoxysuccinates, [23] and Michael acceptors. [24,28,39,40] Serine protease warheads have been based on fluorophosphonates, [8,38] diphenyl phosphonates, [17][18][19] and isocoumarins. [21,22] Threonine proteases react with certain derivatives of vinyl sulfones [25] and epoxy ketones.…”

“…α,β-Unsaturated ketones and their derivatives are precious structures, predominantly distributed in numerous biologically active natural products, medicinal chemicals, agricultural chemicals and functional organic materials [1][2][3]. These structures also serve as starting materials for the synthesis of many valuable organic chemicals [4][5][6].…”
