Evaluation of Three Formulations of Culture Media for Isolation ofBrucellaspp. regarding Their Ability to Inhibit the Growth of Contaminating Organisms

Abstract: Three culture media (Brucella agar, Farrell medium, and CITA) were compared for their effectiveness in inhibiting contamination and for isolating Brucella spp. One hundred lymph nodes from pigs (n = 50) and wild boars (n = 50) with lymphadenitis were collected in slaughterhouses in the State of São Paulo and were assessed on these three selective media for Brucella spp. All of the samples were negative for Brucella spp. on the three culture media. On the agar medium, fungal (70 plates) and Gram-positive bacter… Show more

“…Additionally, this method poses a risk to laboratory personnel and requires specific biosafety measures [22]. Hence, culture and biochemical typing remain the "gold standard" for the diagnosis of Brucella infection [23], including biochemical tests like CO 2 requirement, H 2 S production, and dye sensitivity. Urease, oxidase, and catalase tests are also used for the typing of Brucella spp.…”

“…As a result of these developments, there are a lot of selective media with different basal media, antibiotic mixtures, and concentrations (15). It was emphasized that each medium has a particular effect on the species and biovar of the genus Brucella and contaminants on account of these differences (16,17).…”

“…These factors make brucellosis an urgent problem. Research and development of molecular marked vaccines is an inevitable tendency of research and application of new Brucella vaccines (Vicente et al, 2014). The search for specific protein antigens has gained wide attention.…”

“…The splenocytes and myeloma cells Sp2/0 of the immunized mice were fused using the polyethylene glycol method. Screened positive hybridoma cells were cloned using the limiting dilution method (Vicente et al, 2014). The specific resistance of the hybridoma cell strains obtained was detected by ELISA.…”
