PhilippinesDela Torre, et al.: Antioxidant Capacity of C. pulcherrimaThe study was conducted to determine the antioxidant capacity, expressed as half maximal inhibitory concentration, IC 50 , of Caesalpinia pulcherrima leaf, flower and seed methanol extracts, and their correlation to their total phenolic, flavonoid and triterpenoid contents. Thin layer chromatographic profiling of the methanol extracts was also conducted followed by ultra-performance liquid chromatography quadruple time-of-flight mass spectrometric analysis for the identification of antioxidant compounds. Based on the quantitative antioxidant assays, all extracts exhibited comparable activity with the reference standard at 800 µg/ml (P>0.05). Correlation data revealed a strong negative correlation between the IC 50 and the total phenolic, flavonoid and triterpenoid contents of the extracts, with statistically significant negative correlations observed between the flavonoids of leaf (r=−0.997) and flower (r=−0.998) with reducing power assay, and triterpenoids of flower (r=−1.000) with 2,2-diphenyl-1-picrylhydrazyl scavenging assay. Two common spots with antioxidant activity in the thin layer chromatography profiles were subjected to ultra-performance liquid chromatography quadruple time-of-flight mass spectrometric. The majority of compounds were identified in the library as triterpenoids, flavonoids and phenolics, and still a large quantity of compounds were unidentified. Hyperforin, 3-(4'-hydroxy-benzyl)-5,7-dihydroxy-6,8-dimethyl-chroman-4-one and platycodigenin were identified to be the common compounds present on the three plant parts.

Three polyacetylenes were isolated and purified from Platycodon grandiflorum A. DC for the first time by high-speed counter-current chromatography using a two-phase solvent system composed of hexane/ethyl acetate/methanol/water (1:31:1:31, v/v/v/v) and high-performance liquid chromatography with an Agilent ZORBAX® SB-C18 column (4.6 mm × 150 mm, 5 μm). After separation by high-speed counter-current chromatography and high-performance liquid chromatography, we obtained 3.5 mg of platetyolin A, 4.1 mg of platetyolin B, and 18.1 mg of lobetyolin with purities of 97.2, 96.7, and 96.9%, respectively. The purity of each compound was assessed by high-performance liquid chromatography and the chemical structures were evaluated by high-resolution electrospray ionization time-of-flight mass spectrometry and one- and two-dimensional NMR spectroscopy. Among the isolated compounds, platetyolin A and platetyolin B are newly reported compounds.

Molecularly imprinted polymers were synthesized using mixed tea saponins as a template and acrylamide-β-cyclodextrin as a cofunctional monomer for the specific binding and purification of tea saponins from the defatted cake extract of Camellia oleifera. The adsorption properties of the prepared polymers were systematically evaluated including adsorption kinetics, adsorption isotherms, and selective recognition characteristics. It showed that the adsorption kinetics followed the pseudo first-order kinetic model (R = 0.995) with an equilibrium time of 3 h, adsorption isotherm data fitted well with the Langmuir-Freundlich model (R = 0.984) with an adsorption capacity of 14.23 mg/g. The relative selectivity coefficient (k´) in the presence of the analogues glycyrrhizic acid and glycyrrhetinic acid were 1.16 and 17.21, respectively. The performance of the molecularly imprinted polymers as solid-phase extraction materials was investigated and the results indicated that using acrylamide-β-cyclodextrin as a cofunctional monomer improved both the adsorption capacity and active sites stability of the imprinted polymers. The solid-phase extraction using the polymers as packing materials was subsequently applied for the separation of tea saponins in raw C. oleifera press extract, and targets were obtained with a purity reaching 89%.
