Evaluation of the new Sysmex UF-5000 fluorescence flow cytometry analyser for ruling out bacterial urinary tract infection and for prediction of Gram negative bacteria in urine cultures

Abstract: The new Sysmex UF-5000 showed high diagnostic accuracy in UTI-screening with a very low rate of false negatives. The instrument is capable of predicting Gram negatives with a good SE and a high agreement with the culture, even if this performance needs further evaluation.

“…Overall, the results of both classification methods (LDA and SVM) are very similar, and the prediction accuracy generally exceeds 80%. This compares well with several other methods that demonstrated specificity in the range of 70-85% (Herreros et al, 2015;De Rosa et al, 2018).…”

“…Recent advances in FCM instrumentation have led to the new automated UF-5000 FCM system with higher performance than the device used in the present study [8,9]. Consequently, a more sophisticated workflow is expected in the future because we can reconstruct a regression equation once through multivariate analysis using this new instrument.…”

“…However, FCM alone has limitations in its accuracy. Mass spectrometry has been developed, and adding it into the laboratory workflow has also been reported [8], but this modality is expensive and not practical.…”

“…Flow cytometry has been found to be a promising and fast diagnostic method in patients with infection/sepsis , including urinary tract infections . It may be more reliable than manual counting leukocytes or bacteria in the urine sediment.…”
