Evaluation of the Merlin, Micronaut System for Automated Antimicrobial Susceptibility Testing of Pseudomonas aeruginosa and Burkholderia Species Isolated from Cystic Fibrosis Patients

Abstract: Since accurate antimicrobial susceptibility testing of bacterial cystic fibrosis isolates is known to be problematic and an optimal in vitro testing method has not yet been evaluated, the study presented here was conducted to compare the performance of the reference agar dilution method and broth microdilution with a commercially available automated susceptibility test system (Merlin; Micronaut, Germany). In this pilot study, the susceptibility of 70 clinical strains of Pseudomonas aeruginosa and Burkholderia … Show more

“…For automatic measurement and identification, the SARAMIS™ (spectral archived microbial identification system) database application from bioMérieux SA France was applied. Minimal inhibitory concentrations (MICs) of piperacillin/tazobactam (PIP/TAZ), ceftazidime (CAZ), tobramycin (TOB), amikacin (AMK), ciprofloxacin (CIP) and meropenem (MEM) were determined by the broth microdilution method (Micronaut Merlin) as previously described and interpreted using the criteria recommended by the Clinical and Laboratory Standards Institute (CLSI) [5,6]. Multidrug-resistant isolates were defined as isolates intermediate or resistant to at least three drugs in the following classes: β-lactams, carbapenems, aminoglycosides and fluoroquinolones.…”

“…Therefore, we evaluated the accuracy of the MICRONAUT system for direct AST of positive blood cultures under routine conditions in a clinical microbiology laboratory. The MICRONAUT system is a commercially available, automated, microtiter plate-based broth dilution AST system (2,16). Altogether, 850 positive blood cultures were investigated on a daily basis including weekends during a period of 9 months.…”

“…However, up-to-date studies evaluating newer generations of automated systems in the field of CF microbiology are not available. The Merlin Micronaut system was shown to be reliable for P. aeruginosa CF isolates [39][40]. In 2005 revised CLSI guidelines have been released that for the first time include an approval of the disc diffusion for the testing of B.…”
