Evaluation of the in vitro Anti-inflammatory Activity of Nerium oleander L. Flower Extracts and Activity-Guided Isolation of the Active Constituents

Abstract: Abstract:The in vitro anti-inflammatory activity profile of the Nerium oleander flower EtOH extract/its subextracts (n-hexane, CH2Cl2, EtOAc, remaining H2O) were evaluated on LPS induced Raw 264.7 macrophages. The effects of the crude EtOH extract and its subextracts on nitric oxide (NO) production and cell viability were determined. The most active subextract was determined to be the EtOAc subextract without exerting any toxicity towards Raw 264.7 macrophages. This subextract significantly inhibited NO produc… Show more

“…As mentioned in earlier studies that IL-1β and IL-6 induced during chronic inflammation [30,31,32]. In the present in silico study flavonoids such as Kaempferol and Chlorogenic acid of N. oleander are suitable in relation to binding energy and molecular interaction with amino acids of target receptors when compared to Indomethacin, which is an evidence of experimental study that flavonoids are suitable for anti-inflammation [4]. In other study, a flavonoid…”

“…The flavonoids viz. kaempferol and chlorogenic acid from Nerium oleander and Indomethacin as synthetic ligand were selected as per literature [4]. For all the selected compounds, canonical [23], to determine some specific contacts between the atoms of the test compounds and amino acids of the IL-β and IL-6.…”

“…This pathology, which can also occur as a result of the immune system attacking the synovial membrane, is accompanied by swelling, stiffness, pain, and a reduction or loss of joint function [4]. During the establishment and development of rheumatoid arthritis, many inflammatory mediators play a key role in bone destruction and inflammation of the synovial membrane, including tumor necrosis factor (TNF- α ), interleukin-1 β , interleukin-6, nitric oxide (NO), prostaglandins, reactive oxygen species (ROS), platelet-activating factor, leukotrienes, enzymes (lipoxygenases, cyclooxygenases (COX-1 and COX-2), and phospholipases) [5, 6].…”
