The aim of the present study was to establish the best experimental conditions that lead to the extracts richest in polyphenolic compounds obtained from pomace and canes of Vitis vinifera. In this regard, a D-Optimal design of experiments (DoE) method was applied to investigate the extraction process parameters from each of three materials: red pomace (RP), white pomace (WP) and canes (C). The input variables were the extraction temperature and the ethanol ratio and as response, the total polyphenols content (TPC) was determined. A design space was generated for each of the plant materials and the most concentrated polyphenol extracts were obtained using 50% ethanol at a temperature of 80 °C. Further, the phenolic profiles of the concentrated extracts were detected by LC/MS/MS and the results showed that WP extract was richer in polyphenolic compounds, both flavonoid and phenolic acids, followed by the RP and C extracts. The antioxidant assays revealed that WP and RP extracts exhibited a higher antioxidant activity which correlated to the high content of polyphenols. These findings revealed that RP, WP and C, currently considered agricultural wastes from winery, may be valorized as an important source of natural antioxidants.

Phenolic compound concentrations in Picea abies wood as an indicator of susceptibility towards root pathogens

Quantification of total phenolic compounds in the heartwood of Picea abies using the Folin–Ciocalteu methods showed that concentrations of phenolic compounds in the heart‐ and sapwood of Picea abies varied, depending on the decay class to which boles were allocated. In the heartwood, mean phenolic concentration was 3.28 mg/g, compared with 2.44 mg/g in sapwood. Experiments using different concentrations of phenolic compounds showed that Heterobasidion parviporum is more sensitive to the effects of phenolic compounds found in sapwood, and less sensitive to phenolic compounds found in heartwood. The high concentration of phenolic compounds extracted from spruce stimulated the decomposition of wood by Armillaria ostoyae. This demonstrates the great ability of A. ostoyae to metabolize phenolic compounds. This is confirmed by natural conditions, where A. ostoyae attacks mainly sapwood and H. parviporum attacks heartwood of P. abies. In heartwood, the average antioxidant activity was approximately 10% higher than in the sapwood. The extraction process and presence of amino acids and reducing sugars might influence on the results. The results suggest that timber quality monitoring in spruce stands could be partly based on measurements of phenol compound concentrations in sapwood, as the technique enables prediction of the potential impact of decay fungi early in succession.

Propolis is widely known for its important therapeutic activities. In its chemical constitution, there are several classes of secondary metabolites, such as simple phenols, carboxylic and cinnamic acids, coumarins, tannins and flavonoids. However, flavonoids, an important class of secondary metabolites, may be found in small concentrations in propolis, which makes its quantification difficult. In view of this information, the purpose of this review is to present the main methodologies used in the quantification of this class of secondary metabolites, in addition to proposing new approaches for the characterization of flavonoids and phenols, using low-cost commercial raw materials. A própolis é largamente conhecida por suas importantes atividades terapêuticas. Apresenta em sua constituição química diversas classes de metabólitos secundários, tais como, fenóis simples, ácidos carboxílicos e cinâmicos, cumarinas, taninos e flavonoides. Entretanto, os flavonoides, uma classe importante de metabólicos secundários, muitas vezes encontra-se em pequenas concentrações na própolis, o que pode dificultar sua quantificação. Tendo em vista estas informações, esta revisão teve como objetivo relacionar as principais metodologias utilizadas na identificação e quantificação desta classe de metabólitos secundários, além de propor novas abordagens para a caracterização de flavonoides e fenóis totais, a partir da utilização de matérias primas comerciais de baixo custo.
