Evaluation of the Fluorescence Polarization Assay as a Rapid On-Spot Test for Ruminant Brucellosis in Côte d'Ivoire

Abstract: Brucellosis is a zoonosis of economic and public health concern. While most diagnostic tests for brucellosis can only be performed in the laboratory, the Fluorescence Polarization Assay (FPA) was developed as a rapid point-of-care field test. This pilot project aimed to validate the use of FPA for rapid diagnosis of ruminant brucellosis on the field, and to compare the FPA performance with that of the more commonly used Rose Bengal Test (RBT). Blood samples were first collected from ruminants in a livestock ma… Show more

“…While, there is only a slight kappa agreement between the rapid test and ELISA (κ = 0.18, 95%CI: 0.06–0.31, p = 0.003). However, there is a substantial agreement between the tuberculin tests and ELISA test (κ = 0.72, 95% CI: 0.48–0.95, p < 0.01), which indicates substantial agreement between these two tests [ 36 , 38 ].…”

“…For the cattle data, the study was performed with variables related to animal characteristics and their relations to estimated prevalence against the parallel testing of tuberculin, rapid test, and ELISA. The inter-test agreements among the tuberculin, rapid diagnostic test, and indirect ELISA were calculated by Kappa statistic [ 36 , 38 ].…”

“…Any herd with at least an animal tested seropositive to either RBT or I-ELISA was deemed positive flock. The chi-square test of associations and inter-test agreement between the Rose Bengal Test and ELISA tests for the seropositivity of brucellosis for sheep and goat were calculated [ 34 , 35 ].…”

“…Individual level disease prevalence of sheep and goat and their ock level prevalence were calculated to estimate the overall species-wise disease prevalence in the district. The chi-square test of correlations and inter-test agreement between the Rose Bengal Test and ELISA tests for the seropositivity of brucellosis for sheep and goat were calculated [34][35].…”

“…There was a signi cant association (χ 2 = 28.29, p<0.001) between seropositivity of RBT and ELISA tests for brucellosis in sheep and goats [46] and an extremely high level of agreement (κ = 0.95, 95% CI: 0.85-1, p<0.001 [35].…”
