Evaluation of the Developmental Toxicity Induced by E804 in Zebrafish Embryos

Abstract: E804, a derivative of indirubin, have multi-biological activities such as anticancer and antiinflammatory activities, but little is known about its developmental toxicity. In this study, we investigated the toxicity of E804 on the developments of zebrafish embryos. Our results showed that E804 treatment caused a significant increase of the malformation rate compared with the control groups. Pericardial edema and curved body shape were the most morphological abnormalities observed in E804-treated group. The hat… Show more

“…This can be observed on the increased expression of BAX and Apaf-1 in embryos with glucose 3% exposure. Earlier research studies showed that BAX has an important role in apoptosis process due to ROS, and increased BAX expression correlates with increased apoptosis rate [28][29][30][31]. The result of this study is linear to earlier studies which stated that increased Apaf-1 expression can increase apoptosis rate in zebrafish embryo through mitochondria-dependent pathway [32][33][34].…”

“…The Bcl2 expression involved in the occurrence of apoptosis. Caspase-3 can accelerate the cleavage of several cellular proteins, resulting in the induction of apoptosis . The real-time RT-PCR study revealed that there was no significant difference in PPAR-α, TGF-α, BCL2, and Caspase-3 expression after AgNCs exposure, compared to the control (see Figure S23a in the Supporting Information).…”

Similarly, TGF-α and BCL2 protein expression also showed no significant change (see Figure S23b in the Supporting Information).…”

“…Cebral et al ( 56 ) suggested that H 2 O 2 causes damage to embryos. Wang et al ( 57 ) reported that elevation of ROS promotes the malformation rate of zebrafish embryos. In the present study, exposure to chloroacetamide and its metabolites promoted generation of ROS and increased cell apoptosis in vivo and in vitro .…”
