Evaluation of the antiparkinsonism and neuroprotective effects of hydrogen sulfide in acute 6-hydroxydopamine-induced animal model of Parkinson’s disease: behavioral, histological and biochemical studies

Abstract: Our results show that HS produces significant antiparkinsonism and neuroprotective effects against 6-OHDA neurotoxicity. Since injection of 6-OHDA into MFB produces severe lesion in SN dopaminergic neurons similar to this lesion in the onset of PD in human being, our data recommend HS as potential therapeutic target for treatment of this disease.

“…Our study uses NaHS as the donor of H 2 S, belonging to the follower of the first model which has quite low relevance to a physiological situation. NaHS, an H 2 S donor, was found to protect PC12 cells against MPP~+-induced apoptosis and cytotoxicity [23]. First of all, we found that H 2 S protected SH-SY5Y cells induced by MPP~+ through increasing autophagy flux.…”

“…Motor coordination and balance of rats were tested with rotarod test (Rota-Rod ENV-578(R), MedAssociates) as previously described (24). Briefly, rats were placed on a rotative axis and their latency to fall was measured.…”

“…1 In the last 10 years, a large amount of evidence has confirmed that the neuronal apoptosis of dopaminergic neurons in the substantia nigra is implicated as the main mechanism of neuronal death in the development of PD. 2,3 Developing treatments to slow or prevent the neuronal apoptosis in the substantia nigra of PD is a challenge and a goal worthy of further research.…”

“…4 Recently, emerging research reveals that the inhibition of neuronal apoptosis in the substantia nigra by H 2 S is of great significance in antagonising PD. 2,5,6 However, the mechanism underlying the protection of H 2 S against neuronal apoptosis in PD remains undefined. Autophagy, a process in which lysosomes degrade cell organelles and proteins, is a pivotal participant for the circulation of cellular components, the maintenance of intracellular homeostasis, and the initiation of cell death programs.…”
