Evaluation of the Anti-inflammatory Effects of Atorvastatin on Patients with Rheumatoid Arthritis: A Randomized Clinical Trial

Abstract: BackgroundRheumatoid arthritis (RA) is a chronic inflammatory joint disorder with unknown etiology. Atorvastatin is a lipid-lowering agent that affects the inflammatory processes.ObjectiveThis study aimed to determine the anti-inflammatory effects of atorvastatin on the Disease Activity Index and high-density lipoprotein (HDL) concentrations in RA patients.MethodsThis clinical trial was performed on 38 RA patients, who were referred to the Imam Reza and Ghaem Medical Centers of Mashhad, Iran between 2013 and 2… Show more

“…This disease mainly occurs in individuals aged between 20 and 50 years old and its morbidity rate in women is 2-3 times higher than that in men, particularly in older women. The pathological changes of RA result in functional limitations, working disability and poor quality of life (1-4). One of the important therapeutic approaches for RA in western medicinal therapy is to alleviate pain.…”

“…This persistent inflammatory response is also thought to be an underlying cause for the increased risk of developing CVD in patients with arthritis ( 1 , 2 ). In this context, statins protect from CVD and recent studies suggest that they also reduce disease activity in patients with rheumatoid arthritis ( 4 11 ). It is evident that the pleiotropic actions of statins cannot be uniquely ascribed to inhibition of HMGCA reductase activity.…”

“…These drugs are now widely used in the treatment of CVD as a result of the above effects and because of their limited adverse effects, and in 2005, their sales in the United States was estimated to be $19.7 billion ( 3 ). Recent evidence suggests that statins may also be useful in treating a number of inflammatory diseases, including rheumatoid arthritis, where atorvastatin ( 4 6 ), pravastatin ( 7 ), and simvastatin ( 8 11 ) are suggested to exert joint protective actions. It is now apparent that the protective actions of statins, at least in part, are independent of their ability to regulate HMGCA reductase activity ( 12 ); therefore, during the last few years, there has been significant interest in identifying the mechanisms that are responsible for these beneficial actions.…”

“…Worldwide, 6% of men and 21% of women between 50 and 84 years of age suffer from osteoporosis. Osteoporosis is thus one of the four most common health problems, along with cardiovascular disease, stroke, and cancer 1 2 . The mortality associated with a hip fracture within the first year following occurrence is 8–36% 3 4 .…”
