Evaluation of the anti-hyperglycemic effect and safety of microorganism 1-deoxynojirimycin

Abstract: 1-Deoxynojirimycin (DNJ) is a potent α-glucosidase inhibitor and thus beneficial for prevention of diabetes. While we have succeeded in obtaining the culture supernatant extract (CSE) rich in DNJ from microorganism source, information regarding its anti-hyperglycemic effect and safety were still limited. Therefore, this study was aimed to evaluate the anti-hyperglycemic effect and safety of microorganism DNJ. Oral sucrose tolerance test was performed, and the result showed that CSE was able to significantly su… Show more

“…Previously, we discovered that B. amyloliquefaciens AS385, a bacterial strain that is commonly found in fermented food products, can produce high amount of DNJ after being cultivated in the culture medium supplemented with sorbitol (12). Next, we developed the purification method for B. amyloliquefaciens AS385 culture medium to increase the recovery rate of microbial DNJ (19) by using ethanol precipitation followed by cation exchange absorption-desorption. However, considering the amount of time and cost that are required for this method, simpler method is more preferable for large-scale production.…”

“…When glucose tolerance evaluations are carried out, these are characterized by improved glycemia and a markedly reduced insulinemic response. The α-glycosidase inhibitory activity appears to be superior to that of acarbose and potentially this treatment may reduce the development of diabetes in experimental animals, thus suggesting the application in clinical condition [35].…”

“…1-Deoxynojirimycin (DNJ), an aza-sugar, has structural characteristics similar to those of cyclic monosaccharides; however, the oxygen is substituted with a nitrogen atom. DNJ prevents glucose from entering the bloodstream from the intestines by inhibiting the activities of α-amylase and α-glucosidase [11,12]. Intestinal α-glucosidase is one of the glucosidases of the small intestinal epithelium [13].…”
