Evaluation of the Abbott RealTi m e MTB and RealTi m e MTB INH/RIF Assays for Direct Detection of Mycobacterium tuberculosis Complex and Resistance Markers in Respiratory and Extrapulmonary Specimens

Abstract: bThe Abbott RealTime MTB (RT MTB) assay is a new automated nucleic acid amplification test for the detection of Mycobacterium tuberculosis complex (MTBC) in clinical specimens. In combination with the RealTime MTB INH/RIF (RT MTB INH/RIF) resistance assay, which can be applied to RT MTB-positive specimens as an add-on assay, the tests also indicate the genetic markers of resistance to isoniazid (INH) and rifampin (RIF). We aimed to evaluate the diagnostic sensitivity and specificity of RT MTB using different t… Show more

“…The upgraded version 2.0 software is now available for the RT MTB assay and is recommended for use in conjunction with the RT MTB RIF/INH resistance assay (version 1.0). Recently published studies used the updated software and the direct reflex protocol, and a greater sensitivity was attained (21,22).…”

“…A second‐LPA used for the detection of resistance to anti‐TB drugs includes probes to detect resistance to the class of fluoroquinolones drugs , and the WHO recommended it use to detect resistance in patients diagnosed with resistance to RIF or for the diagnosis of extensively drug‐resistant . Another important molecular technology is the Abbott RealTime MTB RIF/INH resistance, an assay for the detection of RIF and INH resistant MTB in pulmonary specimens which is under evaluation . The main limitations of these assays are that laboratory‐trained staff in PCR, and appropriate laboratory infrastructure is required.…”

“…The cartridge-based XP-MTB additionally identifies Rif-resistance as a marker of MDR-TB [11] and has significantly ameliorated notification of TB and MDR-TB worldwide [12,13]. Now, with the launch of Abbott RealTime MTB and MTB INH/RIF (Abbott, USA), BD MAX™ MDR-TB (BD-MAX, Beckton Dickinson, USA), and Xpert MTB/Rif Ultra (XP-Ultra, Cepheid, USA) (Rif only), a new generation of closed and virtually fully automated qPCR assays detecting DNA of TB bacteria as well as Rif and/or Inh resistance markers are available which, according to the manufacturers', have sensitivities nearing that of culture [14][15][16][17].…”
