Abstract:Two piperazine-containing 3,4-dihyroquinazolines (BK10007S/8S) have been synthesized, based on our previous work on the synthesis and antitumoral activity of 3,4-dihyroquinazolines. After evaluating them for T-type calcium channel blocking effect and in vitro anti-cancer effect, they were profiled for acute and repeat dose toxicity (40 mg/kg, 2 weeks) to BALB/c mice. A new and convenient method for the preparation of 3,4‐dihydroquinazolines 5 with aryl and Ac groups at C(2) and N(3), respectively, has been developed. The key sequence is the formation of aza‐phosphorane intermediates by the reaction of N‐[2‐(azidomethyl)phenyl]benzamides 1 with Ph3P, followed by intramolecular aza‐Wittig reaction and 3‐acetylation, which can be conducted in one‐pot.

This synthetic review has summarized current developments in the preparation of condensed heterocycles containing dihydropyrimidine and tetrahydropyrimidine nuclei by various transformations of preexisting aromatic pyrimidine derivatives. The review presents a variety of strategies for the construction of these highly functional heterocyclic systems.
