Sperm quality in donkeys (Equus asinus) after freezing thawing is still considered lower than that from other animals, including horses. The aim of this study was to test a new freezing extender supplemented with jenny colostrum on donkey sperm. After thawing, we evaluated sperm motility by means of computer-assisted analysis, viability by SYBR-14 and propidium iodide (PI), membrane functional integrity by HOS-test and acrosome integrity by isothiocyanate conjugated with peanut agglutinin (FITC-PNA) and PI. Ejaculates were collected from five fertile Donkeys. Sperm samples were pooled, diluted and cryopreserved into three experimental extender groups: BotuCrio ® , lactose-extender supplemented with egg yolk (20%) and lactose-extender supplemented with jenny colostrum (20%). The results demonstrated that lactose-jenny colostrum samples displayed significantly higher values in almost all parameters evaluated (p < 0.05) compared with the other two extenders after thawing (BotuCrio ® and lactose-egg yolk based extender, respectively)-Total Motility, Viability, HOS test, VCL, VSL and VAP. Acrosome status, LIN, STR and WOB despite showing lower values, none of them were statistically significant (p > 0.05). In conclusion, the extender containing jenny colostrum can be successfully used for donkey semen crypreservation and could effectively improve donkey sperm qualities after freezing-thawing.

Sci., 62, 227−233.The aim of the study was to evaluate the effect of two different extenders on sperm characteristic before equilibration and post-thaw in the endangered Old Kladruber stallions. Also, the response of individual stallions to the extenders used was tested. Semen was collected from six stallions every other day within one week. After centrifugation of the collected sperm-rich fraction, the supernatant was removed and sperm pellets were divided to two aliquots; these were diluted either with Gent (Minitube, Germany) or privately manufactured lactose-EDTA-egg yolk extender (Lact). Three cryopreserved insemination doses (IDs) from each extender (Gent and Lact) were prepared for each stallion from one collection (108 samples from six stallions in total). As a parameter of quality, the motility (computer assisted sperm analysis), viability (fluorescence staining), and morphology (eosin/nigrosine staining) were evaluated after dilution with freezing extenders (fresh) and after thawing (frozen-thawed). The different effects of chosen extenders on the quality of fresh semen were only manifested in higher kinematic parameters of sperm when the Lact extender was used. However, in frozen-thawed samples, the Gent extender yielded significantly better results in all of the evaluated parameters. The representation of sperm subpopulation was significantly influenced by extender in fresh as well as frozen-thawed samples; moreover, we found a significant effect of freezing on the distribution of these subpopulations. The response of individual stallions to chosen extenders was evident in the different quality of fresh as well as frozen-thawed IDs; Gent extender yielded better frozen-thawed IDs. Based on our results, among others describing quality parameters of ejaculate in endangered Old Kladruber stallions, we can recommend using Gent extender for the production of frozen-thawed IDs.

The quality control of frozen semen samples from cattle is established by parameters such as percentage of progressive motility (% MP) because it is related to the fertilization capacity of bulls. Nowadays, sperm quality test is performed by direct visual inspection of sperm through a microscope in andrology laboratories. However, there is a high subjectivity in the observation and assessment depending on the observer; thus, causing unreliable diagnoses and non-repetitiveness in results. The development of a low cost computer tool was proposed to identify the individual sperm motility in cattle through the application of artificial vision algorithms. The methodology consisted of: the acquisition and pre-processing of videos obtained from thawed cryopreserved samples, the segmentation, filtering and detection of spermatozoa using the Fisher Discriminant Analysis and Adaptive Gaussian Models, followed by the assignment and construction of sperm trajectories through the Munkres Algorithm and Kalman Filter. Finally, the characterization and assessment of sperm motility parameters were performed based on the criteria present in computer-aided semen analysis (CASA) commercial systems. The results obtained showed high correlation for the individual sperm motility with a determination coefficient of 0.8143 for 10 different samples of bull sperm with respect to manual analysis. Likewise, a concordance coefficient of 0.966 was found in the 95% confidence interval using the Bland-Altman test, indicating that the measures were highly similar. In this way, the methodology is a reliable technological support that contributes to the improvement of quality control of semen samples from cattle.
