Catabacter hongkongensis was isolated and cultured from human blood for the first time in Scandinavia. The patient, an 83-year-old man from Dalarna, Sweden, recovered without antibiotic treatment, although a high mortality rate associated with C. hongkongensis infection had been reported from China, Canada and France. The genome of the strain ABBA15k was sequenced, assembled and analysed. In contrast to the type strain of the species HKU16T, no antibiotic resistance was observed in Scandinavian strain ABBA15k. The strain was deposited as CCUG 68271, and the draft genome sequence is available from the DNA Data Bank of Japan (DDBJ), the European Molecular Biology Laboratory (EMBL), and GenBank under the accession number LLYX00000000.

Serangan penyakit antraknosa yang disebabkan oleh jamur Colletotrichum gloesporioides pada beberapa tanaman yang bernilai ekonomi tinggi sangat merugikan para petani. Cara konvensional yang banyak dimanfaatkan para petani untuk mengatasi permasalahan tersebut adalah dengan melakukan penyemprotan fungisida sintetik yang berdampak negatif terhadap konsumen dan lingkungan. Alternatif lain yang dibutuhkan untuk menggantikan peran fungisida sintetik tersebut adalah dengan menemukan mikroorganisme antagonis terhadap jamur C. gloesporioides yang kemudian dapat digunakan sebagai bahan biofungisida yang aman bagi kesehatan dan lingkungan. Empat isolat bakteri antagonis jamur C. gloeosporioides yaitu UBCR_12, UBCR_36, UBCF_01, dan UBCF_13 tengah dikembangkan sebagai bahan untuk pembuatan biofungisida. Sebagai tahap awal penelitian pengembangan terhadap keempat bakteri tersebut, dilakukan sejumlah pengujian untuk mengidentifikasi dan mendapatkan sejumlah informasi terkait karakter keempat isolat tersebut. Pengujian morfologi menunjukkan bahwa isolat UBCR_12 adalah , UBCR_36 adalah, UBCF_01 adalah, dan UBCF_13 adalah. Pengujian biokimia menunjukkan bahwa UBCR_12 adalah , UBCR_36 adalah, UBCF_01 adalah, dan UBCF_13 adalah. Sedangkan pengujian dengan molekuler melalui kloning gen 16S rRNA menunjukkan bahwa isolat UBCR_12 adalah Serratia plymuthica.

Simple Staining of Cells on a Chip

Simple staining of cells is a widely used method in basic medical diagnostics, education, and research laboratories. The stains are low-cost, but the extensive consumption results in excessive toxic waste generation. Thus, to decrease the amount of toxic waste resulting from the cell staining procedure is a need. In this study, we developed a magnetically driven and compartmentalized passive microfluidic chip to perform simple staining of human eukaryotic cells, K562 cells, and lymphocyte cells derived from patients. We demonstrated simple staining on cells with trypan blue, methylene blue, crystal violet, and safranin for high, medium, and low cell densities. The stained cells were imaged using a bright field optical microscope and a cell phone to count cells on the focal plane. The staining improved the color signal of the cell by 25-135-pixel intensity changes for the microscopic images. The validity of the protocol was determined using Jurkat and MDA-MB-231 cell lines as negative controls. In order to demonstrate the practicality of the system, lymphocyte cells derived from human blood samples were stained with trypan blue. The color intensity changes in the first and last compartments were analyzed to evaluate the performance of the chip. The developed method is ultra-low cost, significantly reduces the waste generated, and can be integrated with mobile imaging devices in terms of portability. By combining microfabrication technology with cell staining, this study reported a novel contribution to the field of microfluidic biosensors. In the future, we expect to demonstrate the detection of pathogens using this method.
