Evaluation of oxidative stress in mice subjected to aerobic exercise

Abstract: PURPOSE:To evaluate the influence of aerobic exercise on oxidative stress in mice. METHODS:The study included twenty female mice Mus musculus-Swiss divided into two groups: sedentary control (GA) and exercise (GB), each containing ten animals. All animals underwent an adaptation period of seven days isolated in individual boxes. After this period, the animals in the exercise group (GB) were trained in angled running wheel with circumference of 25 cm assembled on an articulated axle during five minutes for thre… Show more

“…Total SOD content was determined spectrophotometrically at 550 nm, and the results were expressed as U·mg −1 protein [ 14 ]. MDA was measured with reference to the MDA assay kit using the thiobarbituric acid method [ 15 ]. The results were expressed as U·mg −1 , and the intensity of the resulting pink color was read at 532 nm.…”

“…The following determinations were made: white blood cell percentage (polymorphonuclear neutrophils (PMN), lymphocytes (Ly), eosinophils (E), monocytes (M), and basophils (B)-described in the Supplementary Material, Figures S4-S6), fibrinogen and C-reactive protein, the activity of liver enzymes: aspartate aminotransferase (AST) and alanine aminotransferase (ALT), kidney function: serum urea and creatinine levels, immune parameters: serum complement (C) [58], serum levels of tumor necrosis factor alpha (TNFα) and interleukin 10 (IL-10), and assessment of oxidative stress through measuring superoxide dismutase (SOD) [59] and malondialdehyde (MDA) activity [60].…”

“…Quantification of plasmatic MDA levels was performed spectrophotometrically based on the thiobarbituric acid method using the RANSOD kit from RANDOX Laboratories Ltd. (Warsaw, Poland) [51]. The activity of GPx in plasma was assessed spectrophotometrically with a specific RANSOD kit from RANDOX Laboratories Ltd., using the reduction procedure of nicotinamide adenine dinucleotide phosphate [52].…”
