Evaluation of Matrix-Assisted Laser Desorption/Ionization Mass spectrometry for second-Generation Lignin Analysis

Abstract: Matrix-Assisted Laser Desorption/Ionization time-of-flight (MALDI-TOF) mass spectrometry is evaluated as an elucidation tool for structural features and molecular weights estimation of some extracted herbaceous lignins. Optimization of analysis conditions, using a typical organic matrix, namely α-cyano-4-hydroxycinnamic acid (CHCA), in combination with α-cyclodextrin, allows efficient ionization of poorly soluble lignin materials and suppression of matrix-related ions background. Analysis of low-mass fragments… Show more

“…Matrix-assisted laser desorption/ionization (MALDI) ToF mass spectrometry has provided another useful tool for herbaceous lignin analysis [73]. Aromatic acids α-cyano-4-hydroxycinnamic acid (CHCA) or 2,5-dihydroxybenzoic acid (2,5-DHB) were found to provide the best MALDI matrices.…”

“…Mass spectrometry, in its numerous configurations, has been employed to characterize the structure of lignin [62,[67][68][69][70][71][72][73][74][75][76][77]. Time-offlight (ToF) secondary ion mass spectrometry (SIMS) is one such MS design that provides enhanced sensitivity to surface measurements of solid analytes with high mass spectral and spatial resolution, requires limited sample preparation, and yields rapid analyses over wide mass ranges [69][70][71][72]74,76].…”

“…In some cases, MALDI was successfully applied to isolated narrow lignin fractions14, 15 but it has not yet evolved into a generally applicable routine method for different types of lignin 16, 17. ESI‐MS also has some inherent problems to grasp lignin as a whole: the analyte cannot be ionized uniformly owing to its rather heterogeneous chemical nature so that usually only smaller fragments are detected, and larger molecules are suppressed.…”
