Evaluation of matrix-assisted laser desorption ionization mass spectrometry for polymer characterization

Abstract: A protocol for the preparation of polymeric samples for time-of-flight matrix-assisted laser desorption ionization mass spectrometry (TOF-MALDI-MS) analysis was developed. Dithranol was identified as a good matrix for polystyrene (PS), and the addition of silver for cationization of molecules was determined to be necessary. Based on this preparative method, low molecular weight samples of other polymers [polyisoprene, polybutadiene, poly(ethylene oxide), poly(methyl methacrylate), and polydimethylsiloxane] wer… Show more

“…The THF solvent was distilled over Na metal to eliminate water which was found to have an adverse effect on the MALDI signal [15]. An optimal MALDI recipe for high mass polystyrene was developed by using a number of preparation methods based on early reports [28,29]. The following combinations of matrix/solvent/cationizing agent were tested: dithranol or all-trans retinoic acid/THF or dichloromethane or toluene/silver nitrate or silver trifluoroacetate.…”

“…Before this study, others have collected mass spectra of high mass polystyrene and PMMA [15,28,29]. The highest reported MALDI TOF distribution of polystyrene ions were observed at m/z 750 kTh for the ϩ2 charge state of the 1.5 MDa polystyrene [15].…”

“…The assumption was valid in both cases, i.e., quantification of oligomers with different end groups by MALDI showed no obvious bias towards a particular end group, and as such no detailed investigation of an end group bias was done. In one case where the influence of end functional groups was quantified, Belu and coworkers explored the relative ionization yields of dimethylphenylsilyl-and perfluoroalkylsilyl-terminated PS as compared to that terminated with a proton but found little difference in the aggregate values [20]. In another study, Weaver and coworkers explored the quantification of hydroxyl-and hydrogen-terminated polystyrene, finding equal yields of the polymers for equal molar amounts.…”

“…This change in peak intensities was attributed to side reactions such as chain redistribution and backbiting. Other studies have also used mass spectrometry to determine structural and mechanistic information on polymeric systems that would not be possible with other techniques [17][18][19][20].…”
