Evaluation of Ionic Liquids and Ionic Liquids Active Pharmaceutical Ingredients Inhibition in Elastase Enzyme Activity

Abstract: Human neutrophil elastase (HNE) is used as diagnostic biomarker for inflammation/infection. In this work, 10 ionic liquids (ILs) and 11 ionic liquids active pharmaceutical ingredients (ILs-APIs) were tested to evaluate the inhibition effect on the activity of porcine pancreatic elastase enzyme, frequently employed as a model for HNE. The insertion of ionic liquids in some drugs is useful, as the insertion of ILs with inhibitory capacity will also slow down all processes in which this enzyme is involved. Theref… Show more

“…Enzyme inhibition assay is considered a good predictor of overall ILs toxicity, and it is widely used for the assessment of the environmental safety of ionic liquids. The methods most commonly referred to are based on acetylcholinesterase (AChE) [ 42 ] and DNA-polymerase enzymes [ 9 ], but also AMP deaminase [ 43 ], monooxygenase P450 [ 44 ] or elastase [ 45 ] based assays have been used.…”

“…Elastase inhibition assay was determined as reported by Mota et al [ 34 ]. First, 50 μL of extract dilutions (31.2 to 1000.0 μg mL −1 ) plus 87.5 μL of HEPES buffer (0.1 M with NaCl 0.5 M, pH 7.5), 10 μL of the substrate (N-succinyl-Ala-Ala-Ala p-nitroanilide, 1.12 mg mL −1 ), 70 μL of acetate buffer (200 mM, pH 5.5) and 2.5 μL of DMSO were added to a 96-well plate.…”
