Green synthesis is based on plant compounds' ability to reduce metal ions and stabilise them into nanoparticles, and it is gaining a lot of attention in recent years. It has been shown to be a reliable and alternate method for the development of novel Nanoparticles. Andrographis echioides (L.) Nees is a used to synthesize zinc oxide nanoparticles. UV-visible spectroscopy, FTIR, Zeta potential, particle size analysis, XRD, SEM, AFM analysis were adopted to characterise the capped ZnONPs. The intense surface Plasmon resonance bands of ZnONPs at 340 nm confirmed the green synthesis of ZnONPs. The various functional groups of A.echioidesinvolved in the synthesis and crystalline nature of ZnO Nanoparticles were identified using FTIR and X-ray diffraction respectively. The average particle size was found to be 4.0 nm, and the zeta potential value was -11 mV. Administration of ZnONPs significantly reduced the blood glucose level and HbA1C when compared with standard drug glibenclamide in STZ- induced in-vivo models. The antidiabetic potential of ZnONPs was confirmed by numerous biochemical parameters like serum lipid profiles, liver and renal functional markers and regeneration β- cells of islets of Langerhans in STZ- induced diabetic rats. Thus, the ZnONPs synthesized from A. echioideswere found to be effective anti-hyperglycemic agent in the management of diabetes.

Background. The leaves of Bersama abyssinica are used for the treatment of diabetes mellitus in folk medicine system of Ethiopia. The present study was done based on the traditional claim of B abyssinica for the treatment of diabetes mellitus. Methods. The α-amylase inhibition and antioxidant activities of B abyssinica extracts were evaluated by using 3,5-dinitrosalicylic acid method and diphenyl-2-picrylhydrazyl assay model, respectively. Blood glucose lowering activity of the extracts was studied in 4 animal models; normoglycemic, oral glucose loaded, and streptozotocin-induced diabetic mice models. Results. Among the extracts, the crude extract showed the highest α-amylase enzyme inhibition activity with an IC50 of 6.57 μg/mL. The water fraction showed the strongest antioxidant activity with an IC50 of 3.43 μg/mL. The crude extract at doses of 200, and 400 mg/kg showed significant ( P < .05) hypoglycemic activity in normoglycemic mice. All doses of the crude extract significantly ( P < .05) reduced blood glucose levels of oral glucose-loaded mice. In streptozotocin-induced diabetic mice models, both the crude and solvent fractions showed a significant ( P < .05) blood glucose lowering effect as compared with the negative control group post 8 hour treatment. Conclusion. The results demonstrated the beneficial biochemical effects of B abyssinica extract by inhibiting α-amylase and scavenging the free radicals. The crude extract and solvent fractions of B abyssinica had significant blood glucose lowering effect in all animal models.

Background: Becium grandiflorum has been used traditionally for treatment of different ailments including diabetes mellitus although it lacks scientific evidence. Thus, the present study was aimed at evaluating the antidiabetic effect of Becium grandiflorum in streptozotocin (STZ)-induced diabetic mice. Methods: The antidiabetic activity of hydro-ethanolic (30:70) leaf extract of Becium grandiflorum was evaluated in STZ (45 mg/kg)-induced diabetic and normal mice. Antihyperglycemic, hypoglycemic, oral glucose tolerance and body weight change effects of the extract were assessed after administering three doses of the extract (200, 400 and 600 mg/kg), glibenclamide 5 mg/kg (reference drug) and 2% Tween 80 (vehicle). One-way analysis of variance and Tukey's post hoc test were used for data analysis. Results: All doses of the extract (200 mg/kg (p<0.05), 400 mg/kg (p<0.05) and 600 mg/kg (p<0.01)) and glibenclamide 5 mg/kg (p<0.001) showed statistically significant blood glucose level reduction in normal mice as compared to Tween 80. The hydroalcoholic extract at a dose of 200 mg/kg (p<0.05), 400 mg/kg (p<0.01) and 600 mg/kg (p<0.001) showed better blood glucose tolerance after 60, 120 and 180-minute treatment duration in normal mice as compared to negative control. In diabetic mice, Becium grandiflorum doses and the reference drug caused maximum reduction in blood glucose level at the end of the 15th day of treatment by 17.61%, 22.52%, 24.62% and 34.12%, respectively. The extract's doses and the standard drug showed significant (p<0.05) improvement in body weight while the diabetic control continued to lose their body weight. Conclusion: Thus, Becium grandiflorum exhibits antihyperglycemic activity in STZinduced diabetic mice, and shows improvement in oral glucose tolerance and body weight, which justifies the claimed use of the plant in ameliorating diabetes mellitus in Ethiopian folk medicine.
