Evaluation of in Vitro Culture Conditions to Demonstrate Pregnancy-Dependent Changes in Luteal Function in the Pig1

Abstract: This study was designed to evaluate the most effective in vitro culture conditions for study of pregnancy-dependent changes in LH-stimulated steroidogenesis in porcine luteal tissue. Mixed luteal cells recovered from the CL of cyclic (Day 12) and pregnant (Day 12 and 16) pigs were incubated either in Eagle's minimum essential medium (MEM) or in MEM supplemented with different concentrations of porcine or calf serum or BSA, or were incubated in a defined medium (DM) containing 2 micrograms/ml insulin, 40 ng/ml … Show more

“…Labelled hormones: (1,2,6,7-3 H) progesterone, (1,2,6,7 (N)-3 H) androst-4-ene-3,17 dione, (1,2,6,7-3 H) testosterone, (2,4,6,7-3 H) oestrone, and (2,4,6,7-3 H) oestradiol were from Amersham, UK. Anti-results reported by Wiesak et al [52] and Baranao and Hammond [5]. Granulosa cells (10 6 viable cells/well) were cultured in 24-well culture plates for 24 h in serum free conditions in a humidified incubator (37 °C) gassed with 95% air and 5% CO 2 .…”

“…Samples in four tubes were immediately frozen for assay of tissue progesterone content before culture (T0 P4). Minced tissue in the remaining tubes was cultured in 2 ml of Eagles minimum essential medium (MEM; Sigma) containing a range of LH concentrations (0, 0.1, 1, 10, and 100 ng pLH/ml), which was previously shown in our laboratory to be effective in assessing luteal cell responsiveness to LH stimulation [29], with four replicates per dose. Luteinizing hormone activity in the media were confirmed by radioimmunoassay (RIA).…”
