Evaluation of fluorescent dyes for measuring intracellular glutathione content in primary cultures of human neurons and neuroblastoma SH‐SY5Y

Abstract: BackgroundReduced glutathione (GSH) protects cells against oxidative injury and maintains a range of vital functions. To study GSH content in human neuronal cell cultures, thiol‐sensitive fluorescent techniques requiring a small number of cells may be of great value, but their GSH specificity has not been established in these cells.MethodsWe tested the efficiency of four currently available GSH fluorescent stains in human neurons and SH‐SY5Y neuroblastoma cells, both cultured in microwells, by using a fluoresc… Show more

“…For detection of GSH, the cells were stained with 10 M MBB. MBB reacts nonenzymatically with GSH and binds to thiol groups, and it is excited by UV light and then emits bluish fluorescence at 450 nm with an intensity that is proportional to the intracellular GSH content (35,36). GSH and GSSG levels were also measured using a glutathione assay kit as per the manufacturer's instructions (37).…”

“…Detection of GSH and Apoptotic Cells-Total intracellular GSH was estimated using monochlorobimane (mBCL), which formed fluorescent adducts with GSH (42,43). Adherent cells were collected and preloaded with mBCl (40 M) in PBS for 10 min, followed by incubation with propyl iodide permeable only to dead cells.…”
