Evaluation of fish sperm quality

Abstract: The development of cheaper technical tools and the availability of well established assay kits provide today the spermatologists with several devices for objective and quantifying assessment of sperm quality. The present paper reviews modern methods on sperm concentration assessment, morphometry, motility processes through flagellum studies, motility objective assessment by computer assisted sperm analysis (CASA), sperm integrity and energetic state measurements.

“…We placed a sample of the ejaculate of one male in the center of a 280-μL water droplet harbored in a concave microscope slide. The droplet was framed with a concentrated glucose solution to provide a dilution gradient attracting sperm toward the edges of the droplet (27). Upon contact with water, sperm were activated and dispersed within the water droplet, and longer-lived sperm were expected to reach the outer edge of the water droplet more frequently than short-lived sperm.…”

“…Therefore, many researchers have still relied on more subjective methods of motility assessment (Rurangwa et al, 2004;Fauvel et al, 2010;Kalbassi et al, 2013). The subjective methods of motility analysis are practical and simple; however, their accuracy depends on the experience of the evaluator and is more prone to human error .…”

“…From a biological standpoint, sperm quality could be defined as the ability ofthe spermatozoa to reach and fertilize the oocyte (Fauvel et al, 2010). In this respect, any quantifiable parameter directly correlated with the fertilization capacity could potentially be used as a sperm quality indicator.…”

“…At this stage, the sperm cells are quiescent but already prepared for accomplishing their fertilizing task (Fauvel et al, 2010). However, sperm activation will be an essential requirement in the pursuit of female gametes, and spermatozoa exploit their swimming ability in order to encounter the oocyte as fast as possible (Cosson et al, 2008a).…”
