Background: Canine distemper virus (CDV) affects both white and grey matter in the central nervous system (CNS). It causes demyelination of white matter tracts, which releases myelin basic protein (MBP) into the cerebrospinal fluid (CSF). MBP levels in CSF could therefore indirectly indicate the extent of white matter damage. The objective of this study was to determine MBP levels in the CSF of dogs infected with natural cases of neurologic distemper. Methods: In this study samples from healthy dogs, clinical cases of CDV and stray dogs from in and around the institute were collected for investigation. The infection of the CNS was established by estimating the albumin quotient and, for 16 cases, by histopathology. CSF biochemical parameters were analysed for 35 sero-positive clinical cases of neurologic distemper. MBP levels in samples were estimated by quantitative ELISA. Stray dogs (6 cases) were also screened for this purpose. Result: MBP levels in CSF were significantly (p≤0.05) increased in natural cases of distemper encephalomyelitis and stray dogs relative to controls. Three of the neurologic distemper cases clinically recovered despite high MBP levels, and two stray dogs also had higher MBP concentrations. High CSF MBP may therefore not be a useful marker for predicting the outcome of neurologic distemper.

SUMMARYVisceral leishmaniasis is a complex disease caused by Leishmania infantum, and in dogs, besides the classical symptoms, there are descriptions of inflammatory alterations in the brain. Brain inflammation is a strictly controlled process, and as the brain counts on the efficiency of the bloodbrain barrier (BBB), we aimed to assess BBB integrity in dogs with spontaneous visceral leishmaniasis. Therefore, we evaluated markers in the cerebrospinal fluid (CSF) and in brain tissue related to BBB disruption and brain inflammation. Elevated albumin quota revealed BBB breakdown, corroborated by increased concentrations of anti-Leishmania antibodies in the CSF. In the brain, albumin and IgG staining formed halos around blood vessels, a classical indicator of BBB leakage. Soluble IgG was also detected in the choroid plexus and ependyma, and in these structures, IgG stained random resident cells. IgG + cells and Fcc-RI + cells were identified in the choroid plexus, ependyma and perivascular in the brain parenchyma. The data support the occurrence of BBB disruption in dogs with spontaneous visceral leishmaniasis, and IgG as a key molecule that is capable of initiating and/or maintaining the inflammatory stimuli in the nervous milieu and the CSF as an important disseminator of inflammatory stimuli within the CNS.

A cinomose é uma enfermidade infectocontagiosa e sistêmica, causada por um vírus da família Paramyxoviridae, do gênero Morbilivírus, que acomete principalmente cães. Sua transmissão pode ocorrer por contato direto e indireto, através de alimentos, aerossóis ou objetos contaminados com secreções contendo o vírus. Comumente, a doença acomete cães jovens e adultos, sem uma predileção racial e sexual. O vírus pode afetar concomitantemente os sistemas respiratório, gastrointestinal, cutâneo e neurológico causando sinais clínicos variáveis, sendo observado com frequência: febre, secreção conjuntival, tosse, pústulas abdominais, secreção nasal purulenta, diarreia muco sanguinolenta, podendo deixar sequelas e reduzindo a qualidade de vida ou até, na maioria dos casos, causar a morte dos animais. A manifestação neurológica da doença é considerada a mais grave e preocupante, pois ocorre a replicação do vírus nos neurônios e nas células da glia, podendo causar necrose na substância cinzenta e também desmielinização na substância branca do cérebro. Ainda não existe um tratamento específico contra o vírus da cinomose, apenas protocolos terapêuticos empregados para o tratamento sintomático e de suporte, sendo de extrema importância a realização de medidas de imunoprofilaxia para prevenção. Frente ao exposto, o presente estudo teve o objetivo de realizar um levantamento bibliográfico à respeito da cinomose em cães, enfatizando a ação do vírus sobre o sistema neurológico na espécie.
