Evaluation of eco‐friendly zwitterionic detergents for enveloped virus inactivation

Abstract: Inclusion of a detergent in protein biotherapeutic purification processes is a simple and very robust method for inactivating enveloped viruses. The detergent Triton X-100 has been used for many years and is part of the production process of several commercial therapeutic proteins. However, recent ecological studies have suggested that Triton X-100 and its break-down products can potentially behave as endocrine disrupters in aquatic organisms, raising concerns from an environmental impact perspective. As such,… Show more

“…Being an enveloped virus, washing hands with water and soap for at least 30 s would be beneficial in deactivating the SARS-CoV-2 [96]. Hand sanitizers can also be used if water and soap are not readily available, while touching eyes, nose, and mouth should be prevented [97].…”

“…These observations have resulted in restrictions on its use through the European Chemicals Agency 5,6 . The pharmaceutical industry as well as other large consumers of TX‐100 ( I ) 7,8 now face the challenge to find an alternative detergent to eventually replace TX‐100 ( I ), and indeed, some alternative detergents have recently been suggested 9–11 …”

“…Restricted usage for well-defined procedures and appropriate waste treatment solutions will most probably still be possible in future. Ecofriendly alternatives such as lauryldimethylamine N-oxide (LDAO) [30] have been shown to exhibit effective virus inactivation in a broad range of protein concentrations but need further evaluation with different viruses and conditions.
