ABSTRACT:This study was done to examine the correlation between differential contents of antioxidants and their antioxidative activities in the seeds obtained from two sorghum cultivars (Nampungchal and Hwanggeumchal) according to harvest time post heading, and to understand the properties of the harvested seeds. The seeds were harvested every 5 days from 25-55 days post heading. Length, width, thickness, and morphological properties of the obtained seeds were increased in proportion to the harvest time. The weights per thousand grains, crude fat, and redness (a * ) clearly changed depending on the harvest time. The grain weight and fat content showed the highest value at harvest time days 45 (HT 45). The seed coat lightness (L * ), crude protein, total polyphenol, flavonoid, and tannin in both cultivars were inversely proportional to the harvest time. In addition, ABTS and DPPH radical scavenging activities were also reduced in inverse proportion to the elapsed harvest time. Antioxidants including polyphenol, flavonoid, and tannin showed positive correlations with the antioxidative activities such as ABTS and DPPH activities, but antioxidants and their activities had no significant correlations between cultivars. We therefore propose that sorghum harvest is performed earlier to improve its antioxidative function.

Sorghum landraces are essential for developing cultivars with improved properties, such as disease tolerance, yield and metabolite content. In this study, 139 genotypes (136 Korean sorghum landraces and 3 control cultivars) collected from various provinces were investigated using eleven agronomical and five biochemical traits. The landraces showed little variation in their qualitative agronomical traits. In contrast, quantitative agronomical and biochemical traits differed significantly among the landraces. It was discovered that 16 landraces matured ahead of all control cultivars. Furthermore, 26 landraces had significantly higher thousand seed weights (TSWs) than two of the control cultivars, including Nampungchal (30.63 g) and Sodamchal (30.53 g), whereas only 1 landrace had a significantly higher TSW than the other control cultivar, Wheatland (37.93 g) (p < 0.05). The levels of total tannin content (TTC), total phenolic content (TPC), 2,2′-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) diammonium (ABTS) radical cation scavenging activity, 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity and ferric reducing antioxidant power (FRAP) were in the ranges of 0.12–428.95 mg CE/g, 1.17–10.23 mg GAE/g, 1.64–67.60 mg TE/g, 0.48–31.99 mg AAE/g and 0.63–21.56 mg AAE/g, respectively, and were all affected by collection area, seed weight and seed color. Landraces from northern provinces were discovered to have higher metabolite contents. Furthermore, large seeds had higher TTC and TPC levels as well as DPPH, ABTS and FRAP activities than medium and small seeds, except for the TTC and FRAP, which were significantly different. In terms of seed color, white seeds had significantly lower metabolite contents and antioxidant activities and were notable in principal component analysis. Correlation analysis revealed positive and significant associations between biochemical traits, as well as between panicle-related agronomic traits. In general, the landraces with superior characteristics could be ideal candidates for sorghum breeding programs.

Comparison of Salinity Tolerance Between Grain and Sweet Sorghum Germplasms [Sorghum Bicolor (L.) Moench]

In order to investigate salt-tolerant sorghum germplasms that can grow in saline soil from newly reclaimed land, we measured a well-established germination rate, and growth characteristics including leaf number, height, and root length in salt-treatment conditions (0.3, 0.6, 0.8, and 1.2%) and untreated control. The highly salt-tolerant sorghum line was confirmed using PCA (principal component analysis) analysis and fuzzy comprehensive evaluation method. Germination rate gradually decreased at doses higher than 0.3%, but the germination rates reached about 70% in IT124115, IS1041, Dansusu4ho, and Dansusu2ho germplasms. At 0.6% salt-treatment condition, the germination rates ranged from 35% to 100%. Only seven germplasms (IT103274, IT101381, IT104110, Dansusu4ho, IS20740, IS22720, and IS27887) had germination rates exceeding 50% at 0.8% salt-treatment. At 1.2% salt-treatment IT124115, IT028385, and IS1041 withered. The total number of leaves decreased similarly for both germplasms at salt levels below 0.6%, and sweet sorghum leaf count was more susceptible than grain sorghum at doses higher than 0.8%. In addition, the height of both germplasms was severely reduced even at low salt concentrations, whereas grain sorghum exhibited a greater sensitivity to salinity stress in terms of root length, while sweet sorghum had longer roots at low concentrations when compared with the untreated control. PCA analysis and fuzzy comprehensive evaluation showed that 29 sorghum accessions could be divided into 3 groups based on the germination rate and morphological traits. Especially, sweet sorghum accessions showed a different pattern of PCA plot when compared with the grain sorghum, and salt tolerance could be divided into 5 groups using MFV in terms of their traits. Taken together, the results from this work will contribute to the development of domestic agriculture utilizing marginal land such as reclaimed land by selecting elite sorghum germplasms that have a high salt tolerance and capacity.
