Background: Many long noncoding RNAs (lncRNAs) have been implicated in general and cell type-specific molecular regulation. Here, we asked what underlies the fundamental basis for the seemingly random appearance of nuclear lncRNA condensates in cells, and we sought compounds that can promote the disintegration of lncRNA condensates in vivo.Results: As a basis for comparing lncRNAs and cellular properties among different cell types, we screened lncRNAs in human pluripotent stem cells (hPSCs) that were differentiated to an atlas of cell lineages. We found that paraspeckles, which form by aggregation of the lncRNA NEAT1, are scaled by the size of the nucleus, and that small DNA-binding molecules promote the disintegration of paraspeckles and other lncRNA condensates. Furthermore, we found that paraspeckles regulate the differentiation of hPSCs.Conclusions: Positive correlation between the size of the nucleus and the number of paraspeckles exist in numerous types of human cells. The tethering and structure of paraspeckles, as well as other lncRNAs, to the genome can be disrupted by small molecules that intercalate in DNA. The structure-function relationship of lncRNAs that regulates stem cell differentiation is likely to be determined by the dynamics of nucleus size and binding site accessibility.

Baculoviral IAP repeat containing 7 (BIRC7/Livin/ML-IAP/KIAP; referred to as Livin throughout the present study) and placental growth factor (PlGF) are not detectable in the majority of normal differentiated tissues, but are present in a number of types of cancer, including hepatocellular carcinoma, ovarian cancer and renal cell carcinoma. The aim of the present study was to assess the expression levels of Livin and PlGF in human osteosarcoma specimens and cell lines, and to analyze the functions of Livin and PIGF in the prognosis of osteosarcoma. The expression levels of Livin and PlGF in 48 osteosarcoma specimens and three osteosarcoma cells were determined using immunohistochemistry and reverse transcription-quantitative polymerase chain reaction. The positivity rates of Livin and PlGF in osteosarcoma specimens were 58.3 and 60.4%, respectively, but were 0% in normal bone tissues. The expression levels of Livin and PlGF were increased in MG-63 cells, compared with those in the other cell lines evaluated in the present study. In addition, the expression levels of Livin and PlGF were significantly associated with tumor diameter and Enneking staging, but were independent of tumor site, age and sex of patients. The expression level of Livin was not associated with PlGF. Furthermore, the 5-year overall survival rate was decreased in the Livin or PlGF expression group, compared with that in the non-expression group (P=0.034 and P=0.012, respectively). The expression levels of Livin and PlGF were independent prognostic factors for patients with osteosarcoma. The results of the present study demonstrated that Livin and PlGF may participate in the pathogenesis of osteosarcoma. Therefore, pharmacological inhibition of Livin or PlGF may provide a novel strategy for osteosarcoma treatment.

A degeneração maligna das lesões da doença de Paget é rara (cerca de 1% dos casos), sendo de mau prognóstico apesar do tratamento. OBJETIVOS: Relatar um caso de sarcoma ósseo secundário da doença de Paget, assim como relatar e discutir a evolução clínica do caso e comparar os dados do caso com os da literatura. MÉTODOS: A amostra foi por conveniência e fez parte dela um paciente do sexo masulino com 72 anos de idade. O critério de inclusão foi possuir o diagnóstico de sarcoma ósseo secundário da doença de Paget comprovado por exames e por avaliação clínica, registrada em prontuário. APRESENTAÇÃO DO CASO: PFS, 72 anos, residente em Itumbiara, portador de hipertensão arterial, sem outras comorbidades relatadas, tabagista e etilista, internado no hospital para tratamento de complicação de doença de Paget, identificada durante a internação anterior. Na presente internação foi verificada aumento volumétrico das áreas sugestiva de transformação maligna sendo submetido a cirurgia que evidenciou sarcoma ósseo secundário a doença de Paget no anatomopatológico. DISCUSSÃO: Classifica-se o osteossarcoma de acordo com o grau de malignidade, podendo ser alto grau ou baixo grau, a sua relação com o osso é classificada como intramedular ou periosteal, sua multiplicidade pode ser solitária ou multifocal, e em primário ou secundário. É importante a associação de terapias adjuvantes, como a quimioterapia, havendo melhora significativa das taxas de cura, sendo importante para os procedimentos de preservação dos membros. Na atualidade, existe uma preocupação maior com a qualidade de vida do paciente após o tratamento oncológico. CONCLUSÃO: A realização deste estudo revelou que o sarcoma ósseo é uma complicação da doença de Paget afetando indivíduos com doença poliostótica de longa data e idosos, o qual o profissional da área da saúde deverá suspeitar dessa doença ao analisar os exames de imagem do paciente, em especial a radiografia.

Abstract: Favorable response to the treatment was obtained with an appropriate multiple disciplinary works in the osteosarcomas of extremities, and there were no considerable side effects and sequels in the long-term follow-up of these treatments.
