Evaluation of BMMSCs-EPCs sheets for repairing alveolar bone defects in ovariectomized rats

Abstract: The aim of this paper is to investigate the effect that bone marrow mesenchymal stem cells (BMMSCs) - endothelial progenitor cells (EPCs), BMMSCs and EPCs sheets have on repairing maxillary alveolar defects in ovariectomized (OVX) rats. In this study, after identification using multi-lineage differentiation and flow cytometry, BMMSCs and EPCs were isolated from female rats. The BMMSCs-EPCs, BMMSCs and EPCs sheets were detected by hematoxylin-eosin (H&E) staining, alkaline phosphatase (ALP) staining and qRT-PCR… Show more

“…The multiple differentiation capacities of PDLSCs and BMSCs were determined according to the methods previously described 35,37. Briefly, 1 × 10 6 cells (P2) were cultured in α-MEM in 6-well plates without inducers until confluence.…”

“…Six immunocompromised mice were sorted into no-ECM + COL4A2 (TCP coated with COL4A2 peptide) and P-dECM + COL4A2 (P-dECM coated with COL4A2 peptide) implant transplantation groups. Briefly, we used a model of maxillary defects in rats as previously reported 37. A defect was prepared with a 3-mm-diameter round bur in the mesial-lingual side of the maxillary first molar in each SD rat, ensuring that the size of each defect was the same as that of the round bur.…”

“…MSCs are widely used in regenerative medicine; they are easily accessible and easy to expand and manipulate in vitro and when transplanted do not evoke a host immune-response [ 16 ]. Although the bone-marrow-derived MSCs are the most used for bone tissue engineering, other sources possess the same proliferation and differentiation abilities typical to be defined as mesenchymal [ 17 ]. Nowadays, new populations of MSCs from oral tissues, including periodontal ligament stem cells (PDLSCs), have been proposed as suitable MSC sources for bone tissue regeneration [ 18 ].…”

“…After 8 weeks post-transfection and in vivo implementation, in rats, the bone gaps were smaller and the tissue had a better developed angiogenic network [85]. The combination of BM-MSCs and endothelial progenitor cells (EPCs) induced more rapid bone formation in case of alveolar bone in vivo on ovariectomized rat [86].…”
