Evaluation of association of common variants in HTR1A and HTR5A with schizophrenia and executive function

Abstract: The 5-HT1A receptor (HTR1A) and the 5-HT5A receptor (HTR5A) are key 5-HT receptors with distinct inhibitory functions. Studies have been conducted to investigate the association of a few HTR1A polymorphisms with schizophrenia, producing conflicting results, and the relationship between HTR5A and schizophrenia has not yet been well investigated. We aimed to examine the association of HTR1A and HTR5A with schizophrenia and executive function. The study included a discovery stage with 1,115 patients and 2,289 con… Show more

“…The HTR1A rs878567 SNP is located in the 3′-UTR of the 5-HT 1A receptor close to the splice site, and is in strong linkage disequilibrium with the rs6295 promoter polymorphism. 61 The rs878567 polymorphism has been associated with major depression, 46,47 schizophrenia, 194,195 psychosis 196 and depression after childhood physical abuse. 197 While many of these associations are similar to those for rs6295, the association with schizophrenia and psychosis was not as robust for rs6295.…”

“…Furthermore, HTR2B is also a drug target (Pitychoutis et al, ). Other HTRs that have been less often investigated, but are nevertheless associated with SZ are HTR1B , HTR3A , HTR3B , HTR7 , and HTR5A , which is associated with patient's cognitive abilities (Pal et al, ; Jajodia, Kaur, et al, ; Gupta et al, ; Guan et al, ; Ikeda et al, ). For example, a SZ‐associated promoter SNP rs1062613 C>T in HTR3A (Jajodia, Kaur, et al, ) may affect DNA methylation and CCCTC‐binding factor binding, as this TF binds more strongly to methylated C allele (Jajodia, Singh, et al, ).…”

“…With the rapid development of high-throughput sequencing, although more and more susceptibility variants of complex diseases have been identified (Guan et al, 2013(Guan et al, , 2014(Guan et al, , 2016a(Guan et al, , 2016b(Guan et al, , 2016cLiu et al, 2014;Zhang et al, 2018a). In recent years, researchers have identified strong association signals between the TLR-9 gene and the risk of KOA in both Han Chinese and Turkish populations (Su et al, 2012a;Balbaloglu et al, 2017;Zheng et al, 2017).…”
