Evaluation of Antioxidant Treatment with Superoxide Dismutase in Rat Liver Transplantation after Warm Ischemia

Abstract: In order to investigate the effects of the exogenously administered radical scavenger superoxide dismutase (SOD) on the orthotopic liver graft, livers from male Wistar rats were transplanted after subjection to 40 min of warm ischemia and 30 min of storage at 4 °C. SOD was given at the onset of ischemia and before reperfusion as a supplement (6,000 IU) to the washout solutions. 30,000 IU were infused into the recipient. SOD reduced tissue levels of thiobarbituric acid-reacting substances at the end of ischemia… Show more

“…Colloids and impermeants prevent extravasation of the preservation solution during washout and perfusion, and therefore form essential components for MP and OMP [50,51,52,53]. Addition of free radical scavengers to clinically applied preservation solutions has been demonstrated to result in an improved efficacy of VSOP and OMP, in particular for the preservation of DCD kidneys and livers [45,54,55,56,57,58]. …”

“…Delayed graft function, PNF, and PDF are directly associated with DCD donation [5,6,29]. DCD grafts are subjected to WI; transplantation of such organ grafts leads to inferior posttransplant outcome and eventually organ failure [7,9]. However, improved preservation techniques have improved DCD graft posttransplant performance [12,30,31].…”

“…Following transplantation, these grafts are prone to higher risk, primary dysfunction (PDF), or primary non-function (PNF). DCD grafts are subjected to warm ischemia (WI); this contributes to the post-transplant outcome, which can lead to increased apoptosis [5,6,7,8,9]. Hence, improved preservation techniques are needed to protect these grafts.…”

“…However, transplantation of organs from these donors is associated with a significantly higher incidence of primary nonfunction, biliary complications and initial liver dysfunction compared with livers from HBD [4,16,17] . ROS have been considered to be involved in liver injury at the procurement, preservation and transplantation from donors without beating hearts [8,18,19] .…”
