Present study was designed to compared the total flavonoids and polyphenols contents and antibacterial activity of hawthorn extracts with different polarities as well as the underlying antibacterial mechanisms. The results showed that among all hawthorn extracts, methanol and ethanol extracts (ME and EE) exhibited high levels of total flavonoids and polyphenols contents, followed by acetone, ethyl acetate, trichloromethane and petroleum ether extracts. ME exhibited the strongest antibacterial activity against tested bacteria, especially Staphylococcus aureus with a 1.25 μg/mL of the minimum inhibitory concentration (MIC) and minimum bactericide concentration (MBC). Further analysis revealed that the main phenolic compounds from ME were epicatechin (281.6 mg/100 g DW), procyanidin B2 (243.5 mg/100 g DW), chlorogenic acid (84.2 mg/100 g DW) and quercetin (78.4 mg/100 g DW). The action mechanism of ME against S. aureus could be ascribed to ME damaging cell wall and cell membrane integrity, inhibiting intracellular enzyme activity, increasing reactive oxygen species (ROS), also changing expression of associated genes and then inducing apoptosis of S. aureus. in addition, the antimicrobial activity of ME against S. aureus has also been demonstrated to be efficient in the food matrix (whole milk). Crataegus pinnatifida Bge. (Rosaceae), informally called hawthorn tree, is a widely cultivated perennial woody plant in China 1. Hawthorn has an extremely long edible history and also a well-known Chinese herbal medicine 2. Hawthorn is rich in carbohydrate, organic acids, vitamins and mineral elements 3 , but also has a range of diverse biological functions including antioxidant, antibacterial effect and pharmacological effects on digestive, cardiovascular and endocrine systems, neuroprotective function and preventing obesity 4-7. Polyphenols have been recognized as one of mainly bioactive compounds in hawthorn, and it contained more than 150 phenolic compounds including procyanidins (procyanidin B2, procyanidin B5 and procyanidin C1), flavonoids (epicatechin, hyperoside, quercetin, rutin and isoquercitrin), and triterpenoids acid (ursolic acid, corosolic acid, oleanolic acid and maslinic acid) 8. Traditionally, hawthorn fruits are consumed directly or processed into jams, jellies, wine, juice and various sweet foods 9. Now, with the booming of functional and health care industry, hawthorn extracts as a kind of nutrient enhancer have been widely used 10,11. Wen et al. studied phenolic contents and cellular antioxidant activity of 80% acetone extracts from three varieties of hawthorn 6. Zhang et al. found the ethyl acetate extracts were only effective in inhibition of LDL oxidation among examined four different solvents 12. The flavonoids are compounds synthesized by plants, and they have properties such as antioxidant, anticancer, anti-inflammatory, and antibacterial, among others. One of the most important bioactive properties of flavonoids is their antioxidant effect. Synthetic antioxidants have side toxic effects whilst natural antioxidants, such as flavonoids from natural sources, have relatively low toxicity. Therefore, it is important to incorporate flavonoids derived from natural sources in several products such as foods, cosmetics, and drugs. For this reason, there is currently a need to extract flavonoids from plant resources. In this review are described the most important parameters involved in the extraction of flavonoids by unconventional methods such as ultrasound, pressurized liquid extraction, mechanochemical, high hydrostatic pressure, supercritical fluid, negative pressure cavitation, intensification of vaporization by decompression to the vacuum, microwave, infrared, pulsed electric field, high-voltage electrical discharges, and enzyme-assisted extraction. There are no unified operation conditions to achieve high yields and purity. Notwithstanding, progress has been achieved in the development of more advanced and environmentally friendly methods of extraction. Although in literature are found important advances, a complete understanding of the extraction process in each of the unconventional techniques is needed to determine the thermodynamic and kinetic mechanisms that govern each of the techniques.

Objective: This study evaluated the phytochemical compounds, antioxidant, and antimicrobial activities of Vernonia amygdalina (VA) leaf extract and used it as an auxiliary herbal constituent in natural hair shampoo. Methods: VA leaf was macerated in various solvents. All crude extracts were phytochemical compounds screened and determined the antioxidant and antimicrobial activities before formulating a natural shampoo formula. Natural shampoos were evaluated with physic-chemical properties and sensory satisfaction. Results: About 95% ethanolic extract was the most suitable substrate for product development. It reveals the inhibition zone from 7.00±0.00 to 15.00±1.00 mm and possesses a broad spectrum of antibacterial agents against both Gram-positive and Gram-negative bacteria. The MICs and minimum bactericidal concentrations value ranged from 6.250 to 100 mg/ml and 25 to >200 mg/ml, respectively. About 95% ethanolic extract also revealed high antioxidant activity. The IC50 value of DPPH of 95% ethanolic extract was 1.88±0.02 μg/ml (% inhibition of 85.73±0.01) and the ferric reducing ability power was 23.00±0.50 mg AAE/100 gDW. These high biological activities may be due to a broad range of phytochemical compounds, including saponins, tannins, flavonoids, terpenoids, steroids, cardiac glycosides, and alkaloids. Conclusions: The VA leaf was suitable to use as an ingredient in natural shampoos with a low detrimental effect on normal skin flora. The shampoo with 0.00188% of 95% ethanolic extract was the most suitable formula in terms of characteristics and stability. This formula also obtained the highest satisfaction level under our sensory evaluation.
